Higher plant population and nitrogen management is an adopted approach for improving crop productivity from limited land resources. Moreover, higher plant density and nitrogen regimes may increase the risk of stalk lodging, which is a consequence of complex interplant competition of individual organs. Here, we aimed to investigate the dynamic change in morphology, chemical compositions and lignin promoting enzymes of the second basal inter-nodes altering lodging risk controlled by planting density and nitrogen levels. A field trial was conducted at the Mengcheng research station (33°9′44″N, 116°32′56″E), Huaibei plain, Anhui province, China. A randomized complete block design was adopted, in which four plant densities, i.e., 180, 240, 300, and 360 × 104 ha−1 and four N levels, i.e., 0, 180, 240, and 300 kg ha−1 were studied. The two popular wheat varieties AnNong0711 and YanNong19 were cultivated. Results revealed that the culm lodging resistance (CLRI) index of the second basal internodes was positively and significantly correlated with light interception, lignin and cellulose content. The lignin and cellulose contents were significantly and positive correlated to light interception. The increased planting density and nitrogen levels declined the lignin and its related enzymes activities. The variety AnNong0711 showed more resistive response to lodging compared to YanNong19. Overall our study found that increased planting densities and nitrogen regimes resulted in poor physical strength and enzymatic activity which enhanced lodging risk in wheat varieties. The current study demonstrated that stem bending strength of the basal internode was significantly positive correlated to grains per spike. The thousand grain weight and grain yield had a positive and significant relationship with stem bending strength of the basal internode. The results suggested that the variety YanNong19 produces higher grain yield (9298 kg ha−1) at density 240 × 104 plants ha−1, and 180 kg ha−1 nitrogen, while AnNong0711 produced higher grain yield (10178.86 kg ha−1) at density 240 × 104 plants ha−1 and with 240 kg ha−1 nitrogen. Moreover, this combination of nitrogen and planting density enhanced the grain yield with better lodging resistance.

In this study, we employed electron microscopy to investigate the cytogenetic and embryologic mechanisms of parthenogenesis induced in the 1BL/1RS male sterile lines of wheat. Analysis of the root tips and acid polyacrylamide gel electrophoresis indicated that all of the male sterile lines and their maintainer lines were 1BL/1RS translocation lines, whereas the restorer lines were non-1BL/1RS translocation lines. Furthermore, the chromosomes of 1BL/1RS wheat lines with T. aestivum cytoplasm and Aegilops cytoplasm (include Ae. kotschyi, Ae. ventricosa, Ae. variabilis) paired abnormally at different rates during meiotic metaphase I (MMI). The translocated segment size of the 1RS chromosome and the specific nuclear–alloplasm interaction impaired the pairing of homologous chromosome in the background of the specific Aegilops cytoplasm at MMI. In addition, the frequency of abnormal chromosomal pairing was directly affected by the frequency of haploid production induced by parthenogenesis. The results of this study provide significant insights into the mechanism of parthenogenesis, which is probably due to the abnormal fertilization of synergid cells in alloplasmic 1BL/1RS wheat.

The aim of this study was to investigate the effects of maternal lead exposure on the learning and memory ability and expression of tau protein phosphorylation (P-tau) and beta amyloid protein (Aβ) in hippocampus of mice offspring. Pb exposure initiated from beginning of gestation to weaning. Pb acetate administered in drinking solutions was dissolved in distilled deionized water at the concentrations of 0.1%, 0.5% and 1% groups. On the 21 th of postnatal day, the learning and memory ability of the mouse pups was tested by Water Maze test and the Pb levels in blood and hippocampus of the offspring were also determined. The expression of P-tau and Aβ in hippocampus was measured by immunohistochemistry and Western blotting. The Pb levels in blood and hippocampus of all exposure groups were significantly higher than that of the control group ( P < 0.05). In Water Maze test, the performances of 0.5% and 1% groups were worse than that of the control group ( P < 0.05). The expression of P-tau and Aβ was increased in Pb exposed groups than that of the control group ( P < 0.05). Tau hyper-phosphorylation and Aβ increase in the hippocampus of pups may contribute to the impairment of learning and memory associated with maternal Pb exposure.

Shewanella putrefaciens supernatant was found to increase the virulence factors of Vibrio parahaemolyticus by efficiently degrading its acylhomoserine lactone (AHL). To further reveal the regulation mechanism and its key degrading enzyme, a potential AHL-degrading enzyme acylase (Aac) from S. putrefaciens was cloned, and the influences of temperature, pH, protein modifiers, and metals on Aac were tested. Aac was significantly influenced by temperature and pH, and exhibited the highest AHL-degrading activity at temperatures of 37 °C and pH of 8. Mg2+ and Fe2+ can further increase the AHL-degrading activity. 10 mM EDTA inhibited its activity possibly by chelating the co-factors (metals) required for Aac activity. Tryptophan and arginine were identified as key components for Aac activity that are critical to its AHL-degrading activity. This study provides useful information on Aac and for V. parahaemolyticus control.

Summary

Formaldehyde in aquatic products was determined by micellar electrokinetic capillary chromatography (MEKC) after derivatization with 2,4-dinitrophenylhydrazine. Separation was carried out at 25 °C and 25 kV, using a fused silica capillary (75 µ internal diameter; 50.5 cm effective length) and an ultraviolet detector set at 360 nm. The optimal background electrolyte was 20 mM sodium tetraborate and 20 mM sodium dodecyl sulfate at pH 9.0 with 3 s hydrodynamic injection at 30 mbar. Electrophoretic analysis took approximately 6.5 min. The correlation coefficient of the calibration curve was 0.999 over the concentration range 2.0–100.0 mg L−1, and the LOD and LOQ values were 0.57 and 1.89 µg mL−1, respectively. The recoveries were from 83.7% to 97.2% with steam distillation as the sample pretreatment method.

Summary

Radix Isatidis has widely useful activities including anti-virus, anti-bacterial. Tryptanthrin, indigo, and indirubin are active ingredients in R. Isatidis. Response surface methodology (RSM)-optimized infrared-assisted extraction (IRAE) was developed and combined with HPLC for simultaneous determination of tryptanthrin, indigo, and indirubin from R. Isatidis. IRAE were investigated through extraction yields of the three components and optimized by RSM. The optimum conditions were as follows: infrared power of 129 W, solid/liquid ratio of 1:40 g/mL, and irradiation time of 22.5 min. IRAE conditions obtained by RSM were not only accurate, but also had practical value reflecting the expected optimization. Subsequently, this novel IRAE method was evaluated by extraction yield of the components of R. Isatidis samples from different regions. Compared with common extraction methods including maceration extraction (ME), reflux extraction (RE), ultrasound-assisted extraction (UAE), and microwave-assisted extraction (MAE), IRAE showed higher yield with advantages of no limitation of solvent selection, low cost, convenience under optimum extraction conditions. These results suggested the potential of RSM-optimized IRAE for extraction and analysis of the water-/fat-soluble compositions of Chinese herbal medicine. A simple chromatographic separation for simultaneous determination of tryptanthrin, indigo, and indirubin from Chinese herbal medicine R. Isatidis was performed on a C18 column (Diamonsil 150 mm × 4.6 mm i.d., 5 μm) with a mobile phase isocratic consisting of methanol and water at a flow-rate of 0.8 mL min−1. The retention times of tryptanthrin, indigo, and indirubin were 15.4, 31.9, and 58.6 min, respectively. The linear equations were obtained as follows: y = −3094.5744 + 21208.792x for tryptanthrin (R = 0.9998, 0.9–18.0 μg mL−1), y = 4730.0448 + 30180.567x for indigo (R = 0.9997, 0.5–10.0 μg mL−1) and y = −6582.9045 + 67069.312x for indirubin (R = 0.9997, 0.4–8.0 μg mL−1). The result showed that RSM-optimized IRAE was a simple, efficient pretreatment method for the analysis of complex matrix.

Patrinia scabra Bunge has long been used in clinic as a traditional Chinese medicine for treating leukemia and cancer and regulating host immune response. Despite their wide use in China, no report on system analysis on their chemical constituents is available so far. The current study was designed to profile the fingerprint of ethyl acetate extract of it, and in addition, to characterize the major fingerprint peaks and determine their quantity. Therefore, a detailed gradient high-performance liquid chromatography was described to separate more than 30 compounds with satisfactory resolution in P. scabra Bunge. Based on the chromatograms of 10 batches samples, a typical high-performance liquid chromatographic (HPLC) fingerprint was established with 23 chromatographic peaks being assigned as common fingerprint peaks. Furthermore, a quadrupole time of flight mass spectrometry (Q-TOF/MS) was coupled for the characterization of major compound. As (+)-nortrachelogenin was the most predominant compound in P. scabra Bunge, the quantification on it was also carried out with the method being validated. As a result, (+)-nortrachelogenin was found to be from 1.33 to 2.21 mg g−1 in this plant material. This rapid and effective analytical method could be employed for quality assessment of P. scabra Bunge, as well as pharmaceutical products containing this herbal material.

A method was developed for the preparative separation of two alkaloids from the crude extract of the radix of Rauvolfia verticillata (Lour.) Baill. in a single run. The two-phase solvent system composed of petroleum ether–ethyl acetate–methanol–water (5:5:2:8, v/v), where triethylamine (40 mmol/L) was added to the upper organic phase as the stationary phase and hydrochloric acid (10 mmol/L) was added to the lower aqueous phase as the mobile phase, was selected for this separation by pH-zone-refining counter-current chromatography (PZRCCC). For the preparative separation, the apparatus was rotated at a speed 850 rpm, while the mobile phase was pumped into the column at 2 mL/min. As a result, 112 mg of reserpine and 21 mg of yohimbine were obtained from 3 g of crude extract in a single run. The analysis of the isolated compounds was determined by high-performance liquid chromatography (HPLC) at 230 nm with purities of over 91.0%, and the chemical identification was carried out by the data of electrospray ionization–mass spectrometry (ESI–MS) and nuclear magnetic resonance (NMR) spectroscopy. The technique introduced in this paper is an efficient method for preparative separation of reserpine and yohimbine from devil pepper radix. It will be beneficial to utilize medicinal materials and also useful for the separation, purification, and pharmacological study of Chinese herbal ingredients.

Rht18, derived from Triticum durum (tetraploid) wheat, is classified as a gibberellic acid (GA)-responsive dwarfing gene. Prior to this study, the responses of Rht18 to exogenous GA on agronomic traits in hexaploid wheat were still unknown. The response of Rht18 to exogenous GA3 on coleoptile length, plant height, yield components and other agronomic traits were investigated using F4:5 and F5:6 hexaploid dwarf lines with Rht18 derived from two crosses between the tetraploid donor Icaro and tall Chinese winter wheat cultivars, Xifeng 20 and Jinmai 47. Applications of exogenous GA3 significantly increased coleoptile length in both lines and their tall parents. Plant height was significantly increased by 21.3 and 10.7% in the GA3-treated dwarf lines of Xifeng 20 and Jinmai 47, respectively. Compared to the untreated dwarf lines, the partitioning of dry matter to ears at anthesis was significantly decreased while the partitioning of dry matter to stems was significantly increased in the GA3-treated dwarf lines. There were no obvious changes in plant height and dry matter partitioning in the GA3-treated tall parents. Exogenous GA3 significantly decreased grain number spike–1 while it increased 1000-kernel weight in both the dwarf lines and tall parents. Thus, applications of exogenous GA3 restored plant height and other agronomic traits of Rht18 dwarf lines to the levels of the tall parents. This study indicated that Rht18 dwarf mutants are GA-deficient lines with impaired GA biosynthesis.

Premature termination codons (PTCs) are an important reason for the silence of highmolecular- weight glutenin subunits in Triticum species. Although the Glu-A1y gene is generally silent in common wheat, we here isolated an expressed Glu-A1y gene containing a PTC, named 1Ay8.3, from Triticum monococcum ssp. monococcum (AmAm, 2n = 2x = 14). Despite the presence of a PTC (TAG) at base pair positions 1879–1881 in the C-terminal coding region, this did not obviously affect 1Ay8.3 expression in seeds. This was demonstrated by the fact that when the PTC TAG of 1Ay8.3 was mutated to the CAG codon, the mutant in Escherichia coli bacterial cells expressed the same subunit as in the seeds. However, in E. coli, 1Ay8.3 containing the PTC expressed a truncated protein with faster electrophoretic mobility than that in seeds, suggesting that PTC translation termination suppression probably occurs in vivo (seeds) but not in vitro (E. coli). This may represent one of only a few reports on the PTC termination suppression phenomenon in genes.
