High ozone (O3) can cause great damage to plants. However, the effect of high O3 on nitrogen (N) absorption, distribution, and utilization in rice at different growth stages under different planting densities is poorly understood. In the present study, a conventional cultivar (Yangdao 6) and a hybrid cultivar (II You 084) with different planting densities were exposed to an elevated amount of O3 (E-O3; 50% higher than that of the control, C-O3) under a freeair gas concentration enrichment (FACE) system. N absorption, distribution, and utilization of the green leaves, stems, and shoots at tillering, jointing heading, and maturity were investigated. Results showed that E-O3 significantly increased the N content in the shoots of Yangdao 6 by 7.5%, 12.7%, and 19.6%, respectively, at jointing, heading, and maturity. Also, the N content in the shoots of II You 084 increased by 5.4%, 6.5%, and 8.4% at the corresponding growth stage upon E-O3 application. E-O3 significantly decreased N accumulation of II You 084 by 8.3%, 4.9%, 4.7%, and 19.2%, respectively, at tillering, jointing, heading, and maturity. Further, E-O3 had a decreasing effect on the N distribution in green leaves (p ≤ 0.05) of both cultivars, but exerted an increasing effect on that in the stems of both cultivars (p ≤ 0.05). In addition, E-O3 significantly decreased the N use efficiency (NUE) for biomass of the two cultivars in all growth stages. These results revealed that E-O3 could increase the N content in rice plants but decrease the N accumulation and utilization in both cultivars. The effects of E-O3 on N absorption, distribution, and utilization were not affected by planting density.
Summary
A simple and rapid method, using online ultraperformance liquid chromatography with photodiode array detection and electrospray ionization mass spectrometry (UPLC-PDA-eλ-ESI-MS/MS), was developed for the in-depth analysis of 50 batches Radix et Rhizoma Rhei. The analysis was performed on a UPLC BEH C18 column using a gradient elution system. Baseline separation could be achieved in less than 7.5 min. At the same time, on the basis of the 50 batches of samples collected from representative cultivated regions, a novel chromatographic fingerprint was devised by UPLC-PDA, in which 27 common peaks were detected and identified by the developed UPLC-MS/MS method step by step according to fragmentation mechanisms, MS/MS data, standards, and relevant literature. Many active components gave prominent [M - H]− ions in the ESI mass spectra. These components include anthraquinones, sennosides, stilbenes, glucose gallates, naphthalenes, and catechins. Furthermore, based on the information of these Radix et Rhizoma Rhei components, and further combined with discriminant analysis, a novel discriminant analysis equation (DAE) was established for the quality control of Radix et Rhizoma Rhei for the first time.
Abstract
Fingernails of pathologically diagnosed normal people, light esophageal epitheliosis patients, severe esophageal epitheliosis patients and esophageal cancer patients were irradiated and their elemental contents were determined by INAA. Multivariate statistical treatment of Ca, Cl, K. Mg, Se and Zn data shows that esophageal cancer patients are distinguishable from non-cancer patients. The accuracy computed by neural networks is greater than 80%.
Abstract
Conducting polyaniline/Cobaltosic oxide (PANI/Co3O4) composites were synthesized for the first time, by in situ deposition technique in the presence of hydrochloric acid (HCl) as a dopant by adding the fine grade powder (an average particle size of approximately 80 nm) of Co3O4 into the polymerization reaction mixture of aniline. The composites obtained were characterized by infrared spectra (IR) and X-ray diffraction (XRD). The composition and the thermal stability of the composites were investigated by TG-DTG. The results suggest that the thermal stability of the composites is higher than that of the pure PANI. The improvement in the thermal stability for the composites is attributed to the interaction between PANI and nano-Co3O4.
Abstract
The principle for the electro-generative simultaneous leaching (EGSL) is applied to simultaneous leaching of pyrite-MnO2 in this paper. A galvanic system for the bio-electro-generative simultaneous leaching (BEGSL) has been set up. The equation of electric quantity vs. time is used to study the effect of produced sulfur on electro-generative efficiency and quantity. It has been shown that the resistance decreased in the presence of Acidithiobacillus thiooxidans (A. thiooxidans) with the increase of electro-generative efficiency. The effects of temperature and grain size on rate of ferrous extraction from pyrite under the conditions of presence and absence of A. thiooxidans were studied, respectively. The changes in the extraction rate of Fe2+ as particle size in presence of A. thiooxidans were more evident than that in the absence, which indicated that the extraction in bio-electro-generative leaching was affected by particle size remarkably. Around the optimum culture temperature for A. thiooxidans, the bigger change in the conversion rate of Fe2+ was depending on temperature. The transferred charge in BEGSL including part of S0 to sulfate group in the presence of (A. thiooxidans) which is called as biologic electric quantity, and the ratio of biologic electric quantity reached to 58.10% in 72 h among the all-transferred charge.
Aegiolops kotschyi cytoplasmic male sterile system often results in part of haploid plants in wheat (Triticum aestivum L.). To elucidate the origin of haploid, 235 wheat microsatellite (SSR) primers were randomly selected and screened for polymorphism between haploid (2n = 3x = 21 ABD) and its parents, male-sterile line YM21 (2n = 6x = 42 AABBDD) and male fertile restorer YM2 (2n = 6x = 42 AABBDD). About 200 SSR markers yielded clear bands from denatured PAGE, of which 180 markers have identifiable amplification patterns, and 20 markers (around 8%) resulted in different amplification products between the haploid and the restorer, YM2. There were no SSR markers that were found to be distinguishable between the haploid and the male sterile line YM21. In addition, different distribution of HMW-GS between endosperm and seedlings from the same seeds further confirmed that the haploid genomes were inherited from the maternal parent. After haploidization, 1.7% and 0.91% of total sites were up- and down-regulated exceeding twofold in the shoot and the root of haploid, respectively, and most of the differentially expressed loci were up/down-regulated about twofold. Out of the sensitive loci in haploid, 94 loci in the shoot, 72 loci in the root can be classified into three functional subdivisions: biological process, cellular component and molecular function, respectively.
Abstract
A microcalorimeter (Setaram c-80) was used to study the thermokinetics of the hydration process of calcium phosphate cement (CPC), a biocompatible biomaterial used in bone repair. The hydration enthalpy was determined to be 35.8 J g–1 at 37.0°C when up to 80 mg CPC was dissolved in 2 mL of citric buffer. In the present study, parameters related to time constants of the calorimeter were obtained by fitting the recorded thermal curves with the function θ=Ae–?t(1– e–?2t). The real thermogenetic curves were then retrieved with Tian function and the transformation rate of the hydration process of CPC was found to follow the equation α=1–[1–(0.0075t)3]3. The microstructures of the hydrated CPC were examined by scanning electron microscopy. The nano-scale flake microstructures are due to crystallization of calcium phosphate and they could contribute to the good biocompatibility and high bioactivity.
In this paper, the wind energy resource in China’s oilfields is analyzed, the difference between China’s energy consumption structure and that of the world in average is analyzed as well, and the application prospect of wind heating technology in China’s oilfields is discussed as an example to reform China’s energy consumption structure. It shows that it is possible to use wind energy as an appropriate heat resource or supplementary heat source in some oilfields to supply heat energy for oil heating and living, more room to improve the energy resource utilization in various fields in China remain. It is a benefit to both oilfield and society to improve energy saving and environments.
Lipopolysaccharide and b-1,3-glucan binding protein (LGBP) is a pattern recognition receptor that can recognize and bind LPS and b-1,3-glucan. LGBP has crucial roles in innate immune defense against Gram-negative bacteria and fungi. In this study, LGBP functions in Portunus trituberculatus innate immunity were analyzed. First, the mRNA expression of PtLGBP in hemocytes, hepatopancreas, and muscle toward three typical pathogen-associated molecular patterns (PAMPs) stimulations were examined using real-time PCR. Results show that the overall trend of relative expressions of the LGBP gene in three tissues is consistent, showing up-down trend. In each group, the highest expression of the LGBP gene was at 3 and 12 h post-injection. The LGBP gene is also expressed significantly higher in the hemocytes and hepatopancreas than in the muscle. The highest level of LGBP was in the lipopolysaccharides (LPS) and glucan-injected group, whereas the lowest level was in the PGN-injected group. Furthermore, bacterial agglutination assay with polyclonal antibody specifically for PtLGBP proved that the recombinant PtLGBP (designated as rPtLGBP) could exhibit obvious agglutination activity toward Gram-negative bacteria Escherichia coli, Vibrio parahaemolyticus, and V. alginolyticus; Gram-positive bacteria Bacillus subtilis; and fungi Saccharomyces cerevisiae. LGBP in Portunus trituberculatus possibly served as a multi-functional PRR. In addition, LGBP is not only involved in the immune response against Gram-negative and fungi, as manifested in other invertebrates, but also has a significant role in anti-Gram-positive bacteria infection.
Molecularly imprinted polymers (MIPs) were synthesized by imprinting a new template—S(-)-1,1′-binaphthalene-2,2′-diamine (S-DABN) and applied as chiral stationary phases for chiral separation of DABN racemates by high-performance liquid chromatography (HPLC). The influence of some key factors on the chiral recognition ability of MIPs, such as the type of functional monomers and porogen and the molar ratio of template to monomer, was systematically investigated. The chromatographic conditions, such as mobile phase composition, sample loading, and flow rate, were also measured. The chiral separation for DABN racemates under the optimum chromatographic conditions by using MIP chiral stationary phase (CSP) of P3, prepared with the S-DABN/MAA ratio = 1/4 and used acetonitrile (2 mL) and chloroform (4 mL) as porogen, showed the highest separation factor (2.14). Frontal analysis was used to evaluate affinity to the target molecule of MIPs. The binding sites (B t) of MIPs and dissociation constant (K d) were estimated as 4.56 μmol g−1 and 1.40 mmol L−1, respectively. In comparison with the previous studies, this approach had the advantages, such as the higher separation factor, easy preparation, and cost-effectiveness, it not only has the value for research but also has a potential in industrial application.
