This research was aimed to study the cell wall degradation and the dynamic changes of Ca2+ and related enzymes in developing aerenchyma of wheat root under waterlogging. An examination of morphological development by light and electron microscope revealed that the structure of cell wall in middle cortical cells remained intact after 12 h of waterlogging and turned thinner after waterlogging for 24 h. At 48 h, the aerenchyma has been formed. The cellulase activity gradually increased in middle cortical cells within 24 h of waterlogging, and decreased with the formation of aerenchyma. Fluorescence detection and subcellular localization of Ca2+ showed the dynamic changing of Ca2+ at the cellular and subcellular levels during the development of aerenchyma. The activity of Ca2+-ATPase enhanced markedly in intercellular space, plasma membrane and tonoplast of some middle cortical cells after 8 h of waterlogging and remained high after 24 h, but it decreased after 48 h of waterlogging. All these suggests that cellulase, Ca2+ and Ca2+-ATPase show a dynamic distribution during the aerenchyma development which associated with the cell wall degradation of middle cortical cells. Moreover, there is a feedback regulation between Ca2+ and Ca2+-ATPase.

Knowledge of the chromosomal distribution of long terminal repeats (LTR) is important for understanding plant chromosome structure, genomic organization and evolution, as well as providing chromosomal landmarks that are useful for chromosome engineering. The aim of this study is to investigate the genomic distribution of Sabrina -like LTR pDbH12, which was first isolated from Dasypyrum breviaristatum (V b genome), on Triticeae species in relation to the genomic evolution and chromosome identification. Fluorescence in situ hybridization (FISH) analysis showed that pDbH12 is present on Dasypyrum (V genome) and Hordeum (H genome) species with the hybridized signals covering the entire chromosomes. However, clone pDbH12 did not hybridize to the genomes of Secale, Triticum, Lophopyrum, Pseduoroengeria, Aegilops, Agropyron desertorum and Elymus. Thinopyrum intermedium displayed fourteen chromosomes that hybridized with pDbH12. Sequential FISH identified these chromosomes as belonging to the J s genome. Results from sequence characterized amplified region (SCAR) marker and dot blot both support the FISH results, and the integrative results suggest that amplification of Sabrina -like LTR retrotransposons is an important factor which involved in the speciation process. Clone pDbH12 could serve as a cytogenetic marker for tracing chromatin from V or V b , H and J s genomes in wheat-alien introgression lines.

Rye (Secale cereale) plays an important role in wheat improvement. Here we report a new triticale, named Fenzhi-1, derived from the wide cross MY11 (Triticum aestivum) × Jingzhou (Secale cereale) after the in vitro rye pollen has been irradiated by He-Ne laser. Morphologically, Fenzhi-1 is characterized by branched-spikes. Genetically, Fenzhi-1 displays stable fertility and immunity to wheat powdery mildew and stripe rust. In situ hybridization (FISH) and seed storage protein electrophoresis revealed that Fenzhi-1 is a new primary hexaploid triticale (AABBRR). The present study not only provides a new method to synthesize an artificial species, but also shows that Fenzhi-1 could be a valuable source for wheat improvement.

Summary

A new HPLC method has been established for determination of 3-monoiodotyrosine (MIT), 3,5-diiodotyrosine (DIT), 3,5-diiodothyronine (T2), 3,3′,5-triiodothyronine (T3), 3,3′,5′-triiodothyronine (rT3), and thyroxine (T4) produced by hydrolysis of iodinated casein with barium hydroxide. The hydrolytic stability of each analyte was evaluated. Iodinated casein was hydrolyzed with saturated barium hydroxide solution for 16 h at 110°C and the barium ions were then removed as barium sulfate. Reversed-phase HPLC was performed on a 2.1 mm × 150 mm, 5 μm particle, C18 column with a mixture of acetonitrile and 0.1% (v/v) formic acid as mobile phase at a flow rate of 0.2 mL min–1. Acetonitrile was maintained at 5% (v/v) for 5 min and then increased linearly to 50% (v/v) within 35 min. All analytes were quantified by measuring the absorbance at 280 nm. Validation data indicated the method was linear, with regression coefficients (R 2) > 0.998, in the concentration ranges investigated. Sensitivity was adequate—limits of detection (LOD) were 0.04–0.38 μg mL–1 and limits of quantification (LOQ) were 0.05–0.38 μg mL–1. Accuracy and precision were acceptable — for all the analytes recovery was 82.0–93.0% and repeatability, as relative standard deviation, was 1.0–3.0%. Hydrolytic stability tests indicated MIT and DIT are much more stable than the other analytes. rT3 was not released directly from iodinated casein but was formed by deiodination of T4 during hydrolysis. The method could be used to identify iodinated casein, to evaluate its activity and quality, and for supervision and regulation of feed additives.

Abstract  

Sixty-two heroin samples were analyzed for their contents of 15 trace elements (Au, Ba, Br, Ca, Ce, Co, Cr, Fe, La, Na, Sb, Sc, Sm, Th, and Zn) by neutron activation analysis (NAA). Large variations of elemental concentrations between samples were found to possess statistical significance. Of all the elements calcium was the most abundant element, followed by zinc and sodium. The concentrations of Au, Ce, Co, La, Sb, Sc, Sm, and Th in all the samples were below 1 mg·g−1. Classification of these heroin samples was achieved by the application of hierarchical cluster analysis. The results show that NAA can provide useful information on the origin of the illicit drugs.

Abstract  

Excess molar enthalpies of binary mixtures for tributyl phosphate (TBP)+methanol/ethanol were measured with a TAM air Isothermal calorimeter at 298.15 K and ambient. The results for xTBP+(1–x)CH3OH are negative in the whole range of composition, while the values for xTBP+(1–x)C2H5OH change from positive values at low x to small negative values at high x. The experimental results have been correlated with the Redlich–Kister polynomial. IR spectra of the mixtures were measured to investigate the effect of hydrogen bonding in the mixture.

Photoinitiating behaviors of oligo(α-aminoketones) (OAK) macrophotoinitiator containing aminoalkylphenone group on free-radical photopolymerization had been investigated by differential photo-calorimetry (DPC). The macrophotoinitiator showed comparative performance with those commercial photoinitiators with lower molecular mass. The effect of photoinitiator concentrations and UV intensity on the polymerization rate was investigated, and the value of exponential factor was found to be 0.5 at the beginning of polymerization, suggesting that the photopolymerization initiated by OAK followed biradical termination mechanism. Photosensitizer isopropyl thioxanthone (ITX) and oxygen severely restricted the polymerization in these systems. Photoinitiators with lower molecular mass showed higher reactivity than those with higher molecular mass.

Abstract  

Solvent extraction of protactinium with tri-iso-octyl-amine (TIOA) in xylene, benzene, carbon tetrachloride and chloroform from HCl, HF, HNO3, HClO4 and H2SO4 media was studied using 233Pa as a radiotracer. The extraction efficiencies of protactinium were determined as a function of shaking time, concentrations of mineral acids in aqueous phase, extractant concentrations and diluents in organic phase. The extraction mechanism was discussed. The results show that the extracted species in the organic phase is [(R3NH)nPa(OH)xCl y 5−xy ].

Doublesex and mab-3-related transcription factor 1 (Dmrt1) is a Z-linked gene that putatively determines the phenotype of gonads in birds. The sex differential expression of Dmrt1 was examined using wholemount in situ hybridization (WISH) in the urogenital systems during embryogenesis. The results revealed that Dmrt1 showed dimorphic expression in chicken gonads, which increased from day 6.5 to day 10.5. The expression of Dmrt1 in male (ZZ) gonads was not twice as much as in female (ZW) gonads, suggesting the existence of other regulatory mechanisms in addition to Z chromosome dosage effect.

Uniformity in the height of main stem and tillers is a key factor affecting ideal plant type, a key component in super high-yielding rice breeding. An understanding of the genetic basis of the panicle layer uniformity may thus contribute to breeding varieties with good plant type and high yield. In the present study, a doubled haploid (DH) population, derived from a cross between indica rice variety Zhai-Ye-Qing 8 (ZYQ8) and japonica rice variety Jing-Xi 17 (JX17) was used to analyze quantitative trait loci (QTL) for panicle layer uniformity related traits. Six, four and three QTL were detected for the highest panicle height (HPH), lowest panicle height (LPH) and panicle layer dis-uniformity (PLD), respectively. qHPH-1-1 and qPLD-1 were located at the same interval on chromosome 1. The JX17 allele(s) of these QTL increased HPH and PLD by 2.57 and 1.26 cm, respectively. Similarly, qPLD-7 and qHPH-7 were located at the same interval on chromosome 7, where the ZYQ8 allele(s) increased HPH and PLD by 3.74 and 1.96 cm, respectively. These four QTL were unfavourable for panicle layer uniformity improvement because a decrease of the PLD was accompanied by decrease of the plant height. qPLD-6 and qLPH-6-1 were located at the same interval on chromosome 6, however here the JX17 allele(s) increased LPH, but decreased PLD, suggesting that this QTL was favourable for improvement of panicle layer uniformity. The markers identified in this study are potential for marker assisted breeding for the improvement of the panicle layer uniformity and ideal plant type.
