In our research we studied the occurrence of the main apple allergen coding gene-families (Mal d 1, Mal d 2, Mal d 3, Mal d 4) in 16 different and most preferably consumed apple varieties. After the DNA isolation by Wizard method the simple PCR reaction was used to examine the apple allergen-coding genes. To identify the presence of the four allergenic protein-coding genes two primer pairs were chosen. The presence of these allergens in most apple varieties could be confirmed. According to our results two varieties — Jonathan and Granny Smith — were found to contain the lowest amount of the coding genes of the allergenic apple proteins studied by us. Besides this, polymorph pattern was obtained by the use of Mal d 1 primer, which may lead to determine apple varieties with small amount of Mal d 1 allergens.The confirmation study of the presence of potential apple allergens by RNA and protein techniques is our plan in the near future.

Abstract

Similarly to other industries wineries also increasingly attempt to minimize and utilize waste to protect our environment. The aim of this study was to determine the optimal parameters (temperature, solvent concentration, and time) of extracting total polyphenol content (TPC) from Tokaji Aszú marc using two different extraction solvents: ethanol–water and isopropanol–water (1:4 solid/liquid ratio). The extractions were achieved based on Central Composite Design with Response Surface Method (CCRD–RSM). The optimal extraction parameters in the case of ethanol–water solvent: 60 °C temperature, 59.5% ethanol concentration in solvent, 5 h. At these parameters the probable TPC concentration is 23966.2 uM GAE/L. The optimal extraction parameters in the case of isopropanol–water solvent: 60 °C temperature, 52% ethanol concentration in solvent, 5 h. At these parameters the probable TPC concentration is 7188.44 uM GAE/L. In both cases the binary solvent was better than the mono-solvent. Ethanol–water solvent was more efficient than the isopropanol–water solvent.

Phosphatase enzymes are capable of releasing phosphate through cleavage of phosphoester bonds. The seasonal importance of this process was examined by using a model substrate paranitrophenylphosphate and the Michaelis-Menten equation to estimate the release rate of PO 4 -P from phosphomonoesters. The seasonal occurrence of phosphomonoesters and acid phosphatase activity was used to estimate the velocity of phosphate release from these compounds. Filter fractionation of phosphatase activity demonstrated that most activity (>60%) was in size fractions less than 0.45 μm. The release rates were highest in May and June (15 to 25 nmol L −1 min −1 ) during the Gonyostomum semen (Raphidophyta) bloom and decreased to less than 2 nmol L −1 min −1 in two weeks and remained low throughout the summer and the fall. Fractionation of 32 P-H 3 PO 4 labelled dissolved organic phosphorus showed this fraction to vary considerably through the year. Potential phosphate release declined through the summer and into the fall. Significance of the co-occurrence of phosphomonoesters and acid phosphatase activity maxima and Gonyostomum semen (Raphidophyta) bloom is discussed.

ICP-OES (inductively coupled plasma optical emission spectrometry) was used to determine the element content of red wines. Development of the method involves various procedures of sample handling as well as repeatability and reproducibility measurements. Measurements were made for the following 16 elements: Al, B, Ba, Ca, Cd, Co, Cr, Cu, Fe, K, Mg, Mn, Na, P, Sr and Zn. The relative standard deviation of repeatability ranged from 0.41% (potassium) to 27% (cadmium) and that of reproducibility varied between 0.73% (boron) to 52% (cobalt). Recovery of the elements was determined by standard addition with results ranging from 90.6% (phosphorus) to 116.2% (boron). After that, 15 red wine samples were measured. Significant differences were found in the Al, B, Ca, Co, Cr, Cu, Fe, K, Mg, Mn, Na, P, Sr and Zn (P<0.001) concentration of wine samples. Correlation studies revealed parallel changes in phosphorus-potassium, phosphorus-magnesium and zinc-manganese concentrations (r=0.857, 0.875, 0.838). According to principal component analysis, measurements of zinc-, sodium-, boron-, copper- and strontium content gave almost the same results as obtained with 16 different elements. According to a two dimensional non-linear map of absolute values of principal component analysis, wines from Szekszárd and Eger could be differentiated, whereas the analysis of wines from Villány, the Mátra Mountains and Lake Balaton showed overlapping results.

A Food and Agriculture Organization of the United Nations Technical Cooperation Programme (TCP)was undertaken on the western corn rootworm (WCR)in 1997 –1998 to establish a permanent moni- toring network,evaluate a containment and control program,test the feasibility and effectiveness of using a Slam ®-based areawide pest management program,develop training materials,and conduct a risk assessment of the potential for WCR spread and establishment in other areas of Europe.TCP countries were Bosnia-Her- zegovina,Croatia,Hungary,and Romania.Bulgaria and Yugoslavia cooperated as unofficial TCP members. The data from the permanent monitoring network showed that the WCR had spread over an area of about 105,600 km 2 in Central Europe and that economic populations had developed on 14,000 km 2 in Yugoslavia through 1998.The containment and control trapping program,although designed to determine the feasibility of restricting the establishment of WCR beetles in an area,did not prove to be successful due to the number of WCR beetles encountered and their rapid movement into previously uninfested areas.The areawide pest management activity showed that the semiochemical Slam was highly efficacious against WCR beetles with residual activity for up to 2 weeks,thus making it a cost-effective alternative to other controls.Also, investigations showed that WCR will continue to spread and establish in other parts of Europe.

Sixty three scale insect species are reported from the Mezőföld area (Hungary), a mosaic area of predominantly intensive agricultural land and scattered grassland and forested areas. In comparison, from the Kőrös-Maros National Park, dominated by natural grassland areas, only 31 species were reported. From the Mezőföld data 4 species were new for the Hungarian fauna and almost all species from Mezőföld were new for the given locality. The Mezőföld fauna could be characterised by more rare species and a lower Global Frequency Value (=higher level of species rarity), suggesting a higher overall conservation value. Scale insect species numbers show a strong negative correlation with the ratio of woody plantations. There was an overwhelming presence of a steppic scale insect species in grassland assemblages, and impoverished woody fauna of the studied wooded areas. However, there was no correlation with plant species number, with the area of natural vegetation, or with the area of loess steppe patches, which shows that the original loess step fauna is impoverished, heavily disturbed. In summary we can say, that the Mezőföld loess scale insect fauna is relatively poor, could be characterised by widely distributed, mezophilous, common species. However, it also has several important steppic elements, and therefore deserves protection.

Among the factors which determine yield reliability an important role is played by disease resistance. One of the breeding aims in the Martonvásár institute is to develop wheat varieties with resistance to major diseases. The winter wheat varieties bred in Martonvásár are examined in artificially inoculated nurseries and greenhouses for resistance to economically important pathogens. The effectiveness of designated genes for resistance to powdery mildew and leaf rust has been monitored over a period of several decades. None of the designated major resistance genes examined in greenhouse tests is able to provide complete resistance to powdery mildew; however, a number of leaf rust resistance genes provide full protection against pathogen attack (Lr9, Lr19, Lr24, Lr25, Lr28 and Lr35). In the course of marker-assisted selection, efficient resistance genes (Lr9, Lr24, Lr25 and Lr29) have been incorporated into Martonvásár wheat varieties. The presence of Lr1, Lr10, Lr26, Lr34 and Lr37 in the Martonvásár gene pool was identified using molecular markers. New sources carrying alien genetic material have been tested for powdery mildew and leaf rust resistance. Valuable Fusarium head blight resistance sources have been identified in populations of old Hungarian wheat varieties. Species causing leaf spots (Pyrenophora tritici-repentis, Septoria tritici and Stagonospora nodorum) have gradually become more frequent over the last two decades. Tests on the resistance of the host plant were begun in Martonvásár four years ago and regular greenhouse tests on seedlings have also been initiated.

Seeds ensure the survival and dispersal of the majority of vascular plant species. Seeds require species-specific germination conditions and display very different germination capacities using different germination methods. Despite the importance of plant generative reproduction, little is known about the germination capacity of the seeds of the Pannonian flora, particularly under field conditions. Our aim was to reduce this knowledge gap by providing original data on the germination capacity of 75 herbaceous species. We reported the germination capacity of 8 species for the first time. We also highlighted the year-to-year differences in the germination capacity of 11 species which could be highly variable between years. The data regarding the germination capacity of target species, as well as weeds and invasive species, can be informative for nature conservation and restoration projects. Our findings support the composition of proper seed mixtures for ecological restoration and also highlight the importance of testing seed germination capacity before sowing.

For understanding local and regional seed dispersal and plant establishment processes and for considering the ecotypes and other forms of specific variability, hard data of locally or regionally measured traits are necessary. We provided newly measured seed weight data of 193 taxa, out of which 24 taxa had not been represented in the SID, LEDA or BiolFlor databases. Our new measurements and formerly published data of locally collected seed weight records together covers over 70% of the Pannonian flora. However, there is still a considerable lack in seed weight data of taxonomically problematic genera, even though they are represented in the Pannonian flora with a relatively high number of species and/or subspecies (e.g. Sorbus, Rosa, Rubus, Crataegus and Hieracium). Our regional database contains very sporadic data on aquatic plants (including also numerous invasive species reported from Hungary and neighbouring countries) and some rare weeds distributed in the southwestern part of the country. These facts indicate the necessity of further seed collection and measurements.
