In this study, the cDNA of homocysteine S-methyltransferase was isolated from Aegilops tauschii Coss., with the gene accordingly designated as AetHMT1. Similar to other methyltransferases, AetHMT1 contains a GGCCR consensus sequence for a possible zinc-binding motif near the C-terminal and a conserved cysteine residue upstream of the zinc-binding motif. Analysis of AetHMT1 uncovered no obvious chloroplast or mitochondrial targeting sequences. We functionally expressed AetHMT1 in Escherichia coli and confirmed its biological activity, as evidenced by a positive HMT enzyme activity of 164.516 ± 17.378 nmol min−1 mg−1 protein when catalyzing the transformation of L-homocysteine. Compared with the bacterium containing the empty vector, E. coli harboring the recombinant AetHMT1 plasmid showed much higher tolerance to selenate and selenite. AetHMT1 transcript amounts in different organs were increased by Na2SeO4 treatment, with roots accumulating higher amounts than stems, old leaves and new leaves. We have therefore successfully isolated HMT1 from Ae. tauschii and characterized the biochemical and physiological functions of the corresponding protein.

The purpose of this study was to evaluate the ability of Lactobacillus rhamnosus to bind patulin (PAT) in the buffer solution and apple juice. The binding of L. rhamnosus to PAT was reversible, which improved the stability of the bacterial complex. The ability to bind PAT can be enhanced with the inactivation of the strain by high temperature and acid treatment. Acid-treated bacteria had the highest PAT binding rate of 72.73±1.05%. The binding rates of acid and high temperature (121 °C) treatments were increased by 21.37% and 19.15%, respectively. L. rhamnosus showed the best detoxification ability to PAT at 37 °C, where the binding rate reached 50.9±1.03%. When the dose of inactivated bacteria powder was 0.02 g ml−1, the minimum concentration of PAT in apple juice was 0.37 µg ml−1. The addition of the L. rhamnosus inactivated powder did not affect the quality of the juice product and effectively bound the PAT in apple juice.

Thioacetamide (TAA) is a potent hepatotoxicant in acute and chronic hepatic injury. The study examined the protective effect of sesame oil against TAA-induced hepatic injury in rats. Hepatic injury was induced by intraperitoneal injection of 100 mg/kg of TAA for 24 h. Triple doses of sesame oil (1, 2, or 4 mL/kg) was given orally 0, 6, and 12 h after TAA treatment. TAA significantly increased serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels. Sesame oil decreased serum AST and ALT levels and significantly inhibited hepatic lipid peroxidation and nitric oxide levels compared with TAA-alone group. Further, sesame oil significantly inhibited TAA-induced hepatic neutrophil activation marker myeloperoxidase activity. However, sesame oil did not affect hepatic tumor necrosis factor, IL-1β and IL-10 generation in TAA-treated group. In conclusion, sesame oil protects against TAA-induced hepatic injury and oxidative stress via the inhibition of neutrophil activation. However, inflammatory cytokines may not be involved in sesame-oil-associated hepatic protection against TAA in rats.

Grains of 12 accessions of Triticum timopheevii (Zhuk.) Zhuk. ssp. timopheevii (AAGG, 2n = 4x = 28) and one bread wheat cultivar Chinese Spring (CS) and one durum wheat cultivar Langdon (LDN) grown across two years were analyzed for grain iron (Fe) and zinc (Zn) concentrations. All the 12 tested T. timopheevii ssp. timopheevii genotypes showed significantly higher concentration of grain Fe and Zn than CS and LDN. Aboundant genetic variability of both the Fe and Zn concentrations was observed among the T. timopheevii ssp. timopheevii accessions, averagely varied from 47.06 to 90.26 mg kg−1 and from 30.05 to 65.91 mg kg−1, respectively. Their grain Fe and Zn concentrations between years exhibited a significantly positive correlation with the correlation coefficients r = 0.895 and r = 0.891, respectively, indicating the highly genetic stability. Flag leaf possessed twice or three times higher concentrations for both Fe and Zn than grain, and a significantly high positive correlation appeared between the two organs with r = 0.648 for Fe and r = 0.957 for Zn concentrations, respectively, suggesting flag leaves might be indirectly used for evaluating grain Zn and Fe contents. Significant correlations occurred between grain Fe and Zn concentrations, and between grain Zn concentration and the two agronomic traits of plant height and number of spikelets per spike. Both the concentrations were not related to seed size or weight as well as NAM-G1 gene, implying the higher grain Fe and Zn concentrations of T. timopheevii ssp. timopheevii species are not ascribed to concentration effects of seed and the genetic control of NAM-G1 gene. There might be some other biological factors impacting the grain’s Zn and Fe concentrations. These results indicated T. timopheevii ssp. timopheevii species might be a promising genetic resource with high Fe and Zn concentrations for the biofortification of current wheat cultivars.

Glutenin subunit alleles at the Glu-D3 locus and their effects on dough properties, pan bread, and dry white Chinese noodle (DWCN) qualities were investigated using 106 winter and facultative wheat cultivars and advanced lines. Allele Glu-D3c (42.5%) was the most frequent glutenin subunit, followed by Glu-D3b (25.5%) and Glu-D3a (23.6%). Glu-D3d and Glu-D3f occurred in only three and six cultivars, respectively. The effect of Glu-D3 was significant forDWCNquality, accounting for up to 16% of the variation, but there were no significant differences between individual Glu-D3 alleles on dough properties and qualities of DWCNand pan bread. Interaction effects Glu-A1 × Glu-D3 and Glu-B1 × Glu-D3 were significant for DWCN quality and loaf volume. More work is needed to understand the effects of Glu-D3 variation on the determination of dough properties and end-use quality.

Abstract

Marine organisms have attracted considerable attention in recent years. In this study, peptides with osteogenic activity from Pinctada martensii were isolated and identified. Additionally, the effects of the hydrolysates on MC3T3-E1 cell proliferation and differentiation were evaluated using the MTT and alkaline phosphatase (ALP) assays, respectively. First, trypsin, pancreatin, and neutral protease were used to hydrolyse the intact shellfish. The hydrolysates with the greatest effects on osteoblast proliferation and ALP activity were separated and purified. Second, fraction WP2 was isolated and purified using a Sephadex G-25 column. WP2, which had the highest osteogenic activity, increased cell growth by 48.57 ± 0.05% and ALP activity by 6.27 ± 0.07 mU. Finally, four novel peptides were identified in WP2 (FDNEGKGKLPEEY, IVLDSGDGVTH, IVLDSGDGVSH, and SSENSDLQRQ) by Orbitrap Fusion Lumos Tribrid orbital liquid chromatography-mass spectrometry. Our findings revealed that P. martensii contains peptides with potential osteogenic activity.

Saccharomyces cerevisiae MERIT.ferm was used as mono- and mixed-cultures with Williopsis saturnus var. mrakii NCYC500 in mango wine fermentation. A ratio of 1:1000 (Saccharomyces:Williopsis) was chosen for mixed-culture fermentation to enable longer persistence of the latter. The monoculture of S. cerevisiae and mixed-culture was able to ferment to dryness with 7.0% and 7.7% ethanol, respectively. The monoculture of W. mrakii produced 1.45% ethanol. The mango wines fermented by S. cerevisiae alone and the mixed-culture were more yeasty and winey, which reflected their higher amounts of fusel alcohols, ethyl esters and medium-chain fatty acids. The mango wine fermented by W. mrakii alone was much less alcoholic, but fruitier, sweeter, which corresponded to its higher levels of acetate esters.

Barley yellow dwarf virus-GAV (BYDV-GAV) is one of the most serious viruses on wheat in China. In this study, five BYDV-GAV isolates collected from five regions in Northwestern China were sequenced. The complete genome sequences generated in this study along with nine genome sequences of BYDV-GAV isolates available in GeneBank were compared and analyzed. The comparative analysis indicated that the complete genomes of BYDV-GAV showed a low level of genetic diversity with nucleotide sequence identities ranging between 97.0% and 99.7%, and the RNA-dependent RNA polymerase gene (ORF1 + ORF2) was the most variable within the complete genome. Phylogenetic analysis indicated that the BYDV-GAV isolates in Northwestern China could be divided into two groups. In addition, two potential recombination events were detected among the 14 BYDV-GAV isolates. This study provided a detailed description of molecular characterization of BYDV-GAV in Northwestern China based on the complete genome sequences, which increased the understanding of genetic diversity of barley yellow dwarf viruses.

Abiotic stresses like salinity and abnormally cold environments cause significant yield losses in many crops including wheat. Therefore, concerted efforts are being made by breeders to develop new varieties with salt and cold tolerance to ensure stable yields over varied environments. This study was undertaken to screen six hundred and seventy-seven accessions of international wheat genetic resources to identify lines with high level of tolerance to salinity and cold environments. Based on the results of two years study in different agroecological locations, 51 accessions were classified as salt tolerant and 115 accessions were classified as cold tolerant. Of these, 35 accessions had good agronomic characteristics. Also, there were 39 genotypes with combined tolerance to cold as well as salinity. Thus, there were good lines which can be used directly or as parents for breeding wheat varieties for wide adaptation and high yield. Further analysis of the data showed that early genotypes had good cold tolerance but a poor salt tolerance. It was also observed that small number of both test spikelet and spikelet, short spike length and good seed-plump were positively associated with cold tolerance. Therefore, maturity and spike traits should be taken into considered when selecting wheat lines for wide adaptation breeding.

Seven Glu-A1 m allelic variants of the Glu-A1 m x genes in Triticum monococcum ssp. monococcum, designated as 1Ax2.1 a , 1Ax2.1 b , 1Ax2.1 c , 1Ax2.1 d , 1Ax2.1 e , 1Ax2.1 f , and 1Ax2.1 g were characterized. Their authenticity was confirmed by successful expression of the coding regions in E. coli, and except for the 1Ax2.1 a with the presence of internal stop codons at position of 313 aa, all correspond to the subunit in seeds. However, all the active six genes had a same DNA size although their encoding subunits showed different molecular weight. Our study indicated that amino acid residue substitutions rather than previously frequently reported insertions/deletions played an important role on the subunit evolution of these Glu-A1 m x alleles. Since variation in the Glu-A1x locus in common wheat is rare, these novel genes at the Glu-A1 m x can be used as candidate genes for further wheat quality improvement.
