Galectin-1 (Gal-1), a mammalian lectin induces apoptosis of T lymphocytes. Contradictory data have resulted in confusing knowledge regarding mechanism of Gal-1 induced T-cell apoptosis. In this paper we aimed to resolve this controversy by comparing cell death induced by low (1.8 μM, lowGal-1) and high (18 μM, highGal-1) concentration of soluble Gal-1. We show that lowGal-1 and highGal-1 trigger phosphatidylserine exposure, generation of rafts and mitochondrial membrane depolarization. In contrast, lowGal-1 but not highGal-1 is dependent on the presence of p56lck and ZAP70 and activates caspase cascade. The results allow the conclusion that the cell-death mechanism strictly depends on the concentration of Gal-1.

Effect of heat stress on the synthesis of soluble heat shock proteins (HSPs) and the regrowth in seminal roots of three cultivated and three wild wheat genotypes was examined. In regrowth experiments, 2-d-old etiolated seedlings were exposed to 23 (control), 32, 35, 37 and 38 °C for 24 h, and 35 and 37 °C (24 h) followed by 50 °C (1 h). The lengths of the seminal roots generally decreased significantly at the end of 48 and 72 h recovery growth periods at 35, 37 and 38 °C temperature treatments compared with control. Genotypic variability was significant level at all temperature treatments for the seminal root length. Also, genotypic differences for the number of seminal roots were determined among the wheat cultivars and between the wild wheat species and the wheat cultivars at all temperature treatments; but genotypic differences among wild wheat species were only detected at 37®50 °C treatment. Acquired thermotolerance for the seminal root length is over 50% at 37®50 °C treatment. The genotypic variability of soluble heat shock proteins in seminal root tissues were analyzed by two-dimensional electrophoresis (2-DE). Total number of low molecular weight (LMW) HSPs was more than intermediate- (IMW) and high- (HMW) HSPs at high temperature treatments. The most of LMW HSPs which were generally of acidic character ranged between 14.2-30.7 kDa. The genotypes had both common (43 HSP spots between at least two genotypes and 23 HSP spots between 37 and 37®50 °C) and genotype-specific (72 HSP spots) LMW HSPs.

Seed protein pattern of control and M 10 mutant soybean ( Glycine max [L.] Merr.) lines in defatted and non-defatted raw flour was studied after 60% 2-propanol extraction, SDS-PAGE separation, colloidal staining and densitometric evaluation to detect a new variant of the protein KTI and/or BBI, furthermore to find new protein(s) of low molecular weight. Electrophoretic separation of defatted and non-defatted control soybean samples showed the same protein patterns. On the densitograms of mutant lines quantitative and qualitative differences could be observed. Defatted raw soy samples reflected more differences in the number of peaks than non-defatted ones. Beside soy trypsin inhibitors, several more soy proteins of low molecular weight are dissolved. KA mutant line 9 has a unique 2-propanol soluble protein pattern, and a new protein band of Rf=0.37 compared to the control line. Sixty percent 2-propanol soluble soybean seed proteins are suitable for cultivar identification and characterization, furthermore to distinguish soybean lines of the same origin.

Separation and analysis of water-soluble proteins (WSP) are important in understanding wheat grain proteome fundamentals. However, due to their high degree of heterogeneity and complexity in the compositions, separating WSP is generally difficult and relevant methodologies are not efficiently developed yet. Capillary electrophoresis (CE) is one of the analytical methods currently used for protein separation and characterization. In the present study, a CE method is established for rapidly separating and characterizing WSP of wheat grains. The established method was tested in various applications including wheat variety and germplasm identification as well as protein synthesis and accumulation studies during different grain development stages subject to genotypic and environmental variations. As results, the characteristic CE patterns of a range of bread wheat cultivars and related species were readily identified. The synthesis and accumulation patterns of wheat WSP during developing grains as well as their stabilities in different environments were also investigated. The technical advancements present in this article appear to be useful for wheat cultivar and germplasm identification as well as genetics and breeding research.

Lycopene content (LC) and soluble solid content (SSC) are important quality indicators for cherry tomatoes. This study attempted simultaneous analysis of inner quality of cherry tomato by Electronic nose (E-nose) using multivariate analysis. E-nose was used for data acquisition, the response signals were regressed by multiple linear regression (MLR) and partial least square regression (PLS) to build predictive models. The performances of the predictive models were tested according to root mean square and correlation coefficient (R2) in the training set and prediction set. The results showed that MLR models were superior to PLS model, with higher value of R2 and lower values of for RMSE firmness, pH, SSC, and LC. Together with MLR, E-nose could be used to obtain firmness, pH, soluble solid and lycopene contents in cherry tomatoes.

Damaged starch, protein and arabinoxylan (AX) content and composition have been related to water absorption (WA) in a large set of samples. We tested 20 modern bread wheat cultivars bred in Hungary, 20 old Hungarian landraces, and 17 cultivars with special biochemical/functional characteristics from all around the world, this last set for international comparison. Grain was field grown in the 2011 and 2012 harvest seasons. Alinear mathematical model has been developed to estimate WA from protein content, starch damage, AX content and the relative amount of soluble proteins with strong correlation (r2 = 0.65) between measured and estimated data. The introduction of a new parameter, related to the cultivar dependent quantitative composition of soluble proteins and determined by lab-on-a-chip (LOC) analysis, largely improved the predictability of WA. Based on the large variation among the level of AX and certain soluble protein components in wheat flour and their significant contribution toWA determination, it was concluded, that these properties could be appropriate target traits to alter them during wheat breeding programs to improve the WA of wheat flour.

Az újonnan kidolgozott forróvizes extrakciós módszerrel (HWP) nyerhető foszfor és kálium vizsgálati eredmények értelmezése érdekében összehasonlító vizsgálatot végeztünk a Magyarországon hivatalos AL módszerrel. A zömében tartamkísérletekből, valamint üzemi táblákból származó 315 talajmintán hasonlítottuk össze a két módszerrel kioldott foszfor- és káliumtartalmakat. Mind a foszfor, mind a kálium esetében a két módszerrel nyert eredmények szoros összefüggésben voltak egymással, de az összefüggések szorosságát a talajok fizikai és kémiai tulajdonságai befolyásolják. A nagy abszolút értékbeli különbségek (átlagosan 10–20-szor nagyobb értékeket ad az AL módszer) ellenére a két módszerrel nyert eredmények – vizsgálati helyenként – a fentiek figyelembe vételével átkonvertálhatók egymásba. További előrelépést jelenthet a két módszer átjárhatóságában, ha még nagyobb számú mintán a foszfor esetében a kalcium-karbonát jelenléte, esetleg a kötöttség, kálium esetében pedig a kötöttség figyelembevételével végezzük el az átszámítást. A vízoldható elemtartalomnak ugyanis – különösen környezetvédelmi szempontból – kiemelt jelentősége van.

Prolamins are alcohol-soluble fraction of cereal proteins involved in immunological response of patients with celiac disease. The aim of this study was to analyse the similar protein complex of selected varieties of cereal, pseudocereal and legume grains by comparison of protein fractions, amino acids composition and SDS-PAGE electrophoresis. The immunoreactivity was tested by Western blot and ELISA methods. ELISA analysis recognizes celiac active epitopes in wheat gliadin (which is reference protein in determination of celiac activity), and also corresponding epitopes in other grain proteins responsible for immunological response of patients with celiac disease. Estimated quantity of celiac active sequences (calculated as gliadin content) below 20 ppm was found in species of amaranth, buckwheat and millet, as well as rice, maize, chickpea and chickling vetch. Immunological reaction with polyclonal antibody was negative for all crops, except oat, maize, millet and foxtail millet.

Dolgozatunkban a hazai Talajvédelmi Információs és Monitoring Rendszerből (TIM) származó mintaanyagban ( n = 629) végzett összehasonlító vizsgálatok eredményeiről számolunk be. A vizsgálatok célja a hagyományos ammónium-laktát-ecetsavban (AL), a 0,01 M kalcium-klorid, valamint a Baker–Amacher extrahálószerekben meghatározott K-tartalom közötti mennyiségi összefüggések és ezeket befolyásoló talajtulajdonságok tanulmányozása, továbbá annak eldöntése, hogy indokolt lehet-e a hagyományos módszer mellett új kivonószerek használata a talajok K-ellátottságának jellemzésére. Az összefüggés-vizsgálatok során az AL-K és a 0,01 M CaCl 2 -K között közepes erősségű korrelációt ( r = 0,76) állapítottunk meg. Számításaink igazolták a korábbi vizsgálatok eredményeit, bizonyították, hogy a két kivonószer a talaj tápelem-tartalékait nem azonos mértékben oldja, illetve cseréli ki. Megállapítottuk, hogy a kötöttség, a pH, a karbonátosság és a humusztartalom módosítja a kivonószerek közötti összefüggés szorosságát. Az AL és a Baker–Amacher kivonószerek összehasonlításakor a tápanyag-ellátottsági kategóriánként meghatározott átlagos dK értékek a javuló ellátottsági kategóriának megfelelően nőttek. Az egyes kategóriákon belül azonban nagy szórást tapasztaltunk, és megállapítottuk, hogy az AL módszerrel jól ellátottnak minősített mintákban is előfordulhat K-lekötődés. Úgy véljük, hogy a tápanyagszükséglet pontos becslésére szükség lehet a hagyományos módszer mellett más kivonószerek alkalmazására is. Az AL mellett, kiegészítő módszerként javasoljuk a 0,01 M kalcium-klorid, valamint a Baker–Amacher kivonószereket, mivel a 0,01 M CaCl 2 -ban oldott kálium mennyisége, vizsgálataink alapján, a növény számára könnyen hozzáférhető kálium mennyiségével arányos, míg a Baker–Amacher kivonószer a lekötődés mértékét jellemzi.
