Summary

The analysis of lipid-phospholipid oxidation products is of primary importance. Although there are established HPLC and LC-MS techniques, it is shown here for the first time that the combination of matrix-assisted laser desorption and ionization time-of-flight (MALDI-TOF) mass spectrometry (MS) and TLC represents a sensitive, fast, and convenient alternative. A mixture of 1-palmitoyl-2-linoleoyl-sn-phosphatidylcholine (PLPC) and -ethanolamine (PLPE) was oxidized under the influence of atmospheric oxygen and characterized by direct positive ion MALDI-TOF MS as well as combined TLC-MALDI. It is shown that much more detailed information — particularly related to the oxidation products of PLPE that have so far been scarcely investigated — can be obtained by TLC-MALDI. However, it is also shown that further methodological improvements are necessary to make this method generally applicable to complex lipid mixtures.

Abstract  

The thermal decomposition of methano-fullerene derivatives such as ethoxycarbonyl methano[60] fullerene and various isomers of bis-(ethoxycarbonyl methano)[60] fullerene leads to new fullerene derivatives, which have been preliminary characterized. The analysis of separated species was performed by UV-VIS, IR, H- and C-NMR, STM, FAB, LDI and MALDI-TOF MS spectroscopy. One of the isolated phases is a C122 molecule with a dumbbell-like structure.

Abstract  

Matrix-assisted laser desorption/ionization coupled with time-of-flight mass spectrometry (MALDI/TOF-MS) was used for the analysis of low-molecular phosphate compounds found in Hanford tank wastes. The mass spectra of these compounds indicate protonated peaks as well as sodium adducts. Analytical methods presently utilized for the analysis of the phosphate-related organics are both time consuming and labor intensive. A promising alternative is MALDI/TOFMS. The MALDI process produces both positive and negative ions directly and very little sample is required. In addition, there is limited sample preparation and minimal hazardous waste production.

In this study, outer membrane proteins (OMPs) of colistin-resistant Klebsiella pneumoniae and Enterobacter asburiae were analyzed. One colistin-susceptible and three colistin-resistant K. pneumoniae sequence type 258 strains as well as one colistin-susceptible E. asburiae and its colistin-heteroresistant counterpart strain were involved in the study. OMP analysis of each strain was performed by microchip method. Matrix-assisted laser desorption ionization time of flight/mass spectrometry (MALDI-TOF/MS) investigation was carried out after separation of OMPs by two-dimensional gel electrophoresis and in-gel digestion. The MALDI-TOF/MS analysis of OMPs in the colistin-susceptible K. pneumoniae found 16 kDa proteins belonging to the LysM domain/BON superfamily, as well as DNA starvation proteins, whereas OmpX and OmpW were detected in the colistin-resistant counterpart strains. OmpC and OmpW were detected in the colistin-susceptible E. asburiae, whereas OmpA and OmpX were identified in the colistin-resistant counterpart. This study demonstrated that OMP differences were between colistin-susceptible and -resistant counterpart strains. The altered Gram-negative cell wall may contribute to acquired colistin resistance in Enterobacteriaceae.

The characterization of the old Hungarian varieties and landraces is an important part of Hungarian cereal research and breeding. Analysis of these germplasms with the most up-to-date methodologies results a broad scale of diversity of glutenin alleles, which proves their genetic heterogenicity. Exploitation of this attribute is an untapped possibility for developing modern varieties in our breeding programs. The previous research work revealed this diversity by SDS-PAGE analysis and MALDI-TOF technology. The powerful tool, the high throughput lab-on-a chip technique can facilitate the effectiveness of this function and decreases the cost of the analysis. This study demonstrates the application of this technique for analysing the old varieties. The allelic composition and their effects on bread making quality concluded by means of functional analysis.

The genus Rhizobium colonizes in leguminous plants by symbiotic relationship and enriches soil nitrogen through the formation of the root nodule. Legumes are the appropriate crops for the recovery of marginal lands and they can easily grow in adverse climatic condition. Rhizobium sp. VMA301 was isolated from the root nodules of V. mungo , grown in arsenic contaminated field. The LC 50 value of arsenite for VMA301 was found to be 1.8 mM. Sixteen differentially expressed proteins were identified using RP-HPLC and MALDI ToF mass spectrometry from arsenite induced whole cell lysate soluble proteins. It is also found that nine proteins were up-regulated and seven proteins were down-regulated in comparison to the control group (cells grown without arsenite). These differential protein expressions mitigate the toxic effect of arsenite and stimulate the detoxification process.

Absztrakt:

A Fusarium-, Acremonium- és Sarocladium-gombafajok nevezéktana 2017-ben megváltozott. A morfológiailag homogén, de filogenetikailag heterogén fajokat, illetve fajkomplexeket ma már nukleinsav-összetételük szerint a MALDI–TOF MS-vizsgálat segítségével el lehet különíteni. Ez indokolta a taxonómiai besorolás módosítását. Közleményünkben a Fusarium- és Sarocladium-fajok okozta szaruhártya-gyulladás (keratitis) klinikai lefolyását ismertetjük, összefoglaljuk a diagnosztikus és kezelési lehetőségeket. Mindezek jelentős kihívást jelentenek a szemorvos számára. Súlyos szövődményekhez vezet a késői felismerés és kezelés, a folyamat gyors progressziója, a kórokozó átjutása a Descemet-membránon, a gombaellenes terápia korlátozott hozzáférhetősége és penetrációja, valamint a gyakori terápiarezisztencia. A diagnózis felállításához az anamnézisben szereplő trauma vagy kontaktlencse-viselés, PCR és MALDI–TOF MS-vizsgálat, konfokális mikroszkópia, a minta mikrobiológiai tenyésztése és a gyulladás területéből vett citológiai minta mikroszkópos vizsgálata vezet. A primeren választandó konzervatív terápia a lokálisan alkalmazott 5%-os natamicin, illetve esetenként érzékenység meghatározását követően az 1%-os vorikonazol vagy 0,15–0,25%-os amfotericin B szemcsepp, ezenkívül a 0,02%-os polihexametilén-biguanid (PHMB) szemcsepp használatának sikeréről is beszámoltak. Fusarium okozta keratitisben napi 2 × 200 mg vorikonazol tabletta adása is javasolt lehet. Terápiarezisztens esetekben korai, széles átmérőjű perforáló keratoplasztika (PKP) végzendő, épben történő trepanációval. Megkésett diagnózis és specifikus kezelés esetén a gombafonalaknak a Descemet-membránon történt átjutásával az esetek mintegy negyedében a látás elvesztése és a szemgolyó eltávolítása következik be. Jelen közleményünk öt, Fusarium- és Sarocladium-fajok okozta keratitis klinikai lefolyásának változatosságát is bemutatja. Orv Hetil. 2019; 160(1): 2–11.
