Abstract

Recombinant antibody fragments like single chain variable fragments (scFvs) represent an attractive yet powerful alternative to immunoglobulins and hold great potential in the development of clinical diagnostic/therapeutic reagents. Structurally, scFvs are the smallest antibody fragments capable of retaining the antigen-binding capacity of whole antibodies and are composed of an immunoglobulin (Ig) variable light (VL) and variable heavy (VH) chain joined by a flexible polypeptide linker. In the present study, we constructed a scFv against bovine IgA from a hybridoma cell line IL-A71 that secretes a monoclonal antibody against bovine IgA using recombinant DNA technology. The scFv was expressed in Escherichia coli and purified using immobilized metal affinity chromatography (IMAC). The binding activity and specificity of the scFv was established by its non-reactivity toward other classes of immunoglobulins as determined by enzyme-linked immunosorbent assay (ELISA) and immunoblot analysis. Kinetic measurement of the scFv indicated that the recombinant antibody fragment had an affinity in picomolar range toward purified IgA. Furthermore, the scFv was used to develop a sensitive ELISA for the detection of foot and mouth disease virus (FMDV) carrier animals.

Recent research on L2 acquisition has been stressing the potential of audiovisual translation as a tool for boosting foreign language competence. Whereas most studies have concentrated on subtitled input, less attention has been devoted to dubbing, which is nevertheless the main audiovisual translation modality in several countries. Being the outcome of a translation process, dubbed dialogue is subject to translation universals, including simplification, explicitation and standardisation. These strategies may contribute to the greater accessibility of dubbed vis-à-vis original, non-translated products to non-native viewers. With a view to exploring the role of dubbing in ultimately fostering second language acquisition, an empirical study on the comprehension of different types of audiovisual input by learner-viewers was designed. The study moves from the assumption that input comprehension is a necessary prerequisite for acquisition proper and compares the degree of comprehension of dubbed vs. non-translated film scenes among intermediate-level learners of L2 Italian. Quantitatively and qualitatively comparable film scenes were selected and dialogue comprehension was assessed through closed and timed questions administered after exposure to each scene. Findings show that dubbed audiovisual input results in better comprehension than non-translated film dialogue independently of viewers’ L1 and audiovisual texts’ individual features. The study thus paves the way for further research on the acquisitional impact of dubbed dialogue, especially among learners at lower proficiency levels.

Invertebrate microglia constitute a class of cells resident in the ganglionic nervous system which are activated after tissue injury or by the presence of pathogens. The microglia activation response includes graduated morpho-functional and biochemical changes and cell proliferation. In this study we verified in the freshwater snail Planorbarius corneus that an activation caused by a traumatic event may induce microglia division. Cell proliferation was assessed immunocytochemically using BrdU incorporation technique and documented on both ganglionic sections and microglia cultured cells at different experimental conditions and times after activation. In addition, we studied the possibility of increasing microglia proliferation by adding to the cultured medium the Macrophage-Colony Stimulating Factor (M-CSF) that has been shown to stimulate specifically this process in vertebrates.

The imperative of integrating empirical and theoretical considerations when developing policy responses to Internet-gaming disorder

This commentary proposes an integrative approach of theoretical and empirical considerations when developing policy responses to Internet-gaming disorder and when evaluating their efficacy. It complements the overview by Király et al. (2018) about preventive and treatment programs by referring to a lack of inclusion of internal factors, such as individual aspects and cognitions, and missing empirical evidence. This commentary claims the integration of current research addressing individual predisposing and maintaining factors in order to evaluate existing programs and to enhance the exchange between actors including policy. This integrative approach has the potential to develop successful preventive strategies, which could be implemented realistically and socially responsible.

Summary

This paper investigates some features of the technical legal terms in Leges Langobardorum. Rothair’s Edict was the first written codification of the Lombard customary law. The passage into writing, the writers’ choice of Latin and the use of Roman law texts as sources and models play an important role in determining the features of their legal vocabulary. Within this perspective, the specialized lexemes of Lombard Laws, the use of terms in ordinary and technical meanings and the polysemy and monoreferentiality of legal terminology are discussed.

The effect of heat induced changes on the immunoreactivity of a-lactalbumin (a-la) in whey was evaluated. Whey and acidified a-la solutions were heated for various times at temperatures between 60 and 100 °C. After heating, the samples were clarified and the protein content (by Bradford's method), the quantity of a-la (by FPLC method) and the immune response to an anti a-la probe were determined. The samples heated at 80 °C were characterized by the highest a-la immunoreactivity properties. Raising temperature up to 90 °C and 100 °C caused a marked decrease in a-la immunoreactivity. As was found by FPLC method in the samples heated at 60 and 65 °C, a-la content changes were not statistically significant. A gradual decrease in the content of a-la was observed with increased temperature and duration of heating.

The aim of this study was to investigate differences between the progesterone profiles of Beagle bitches with (multiparous) and without (nulliparous) the whelping experience and to examine whether the selection of bitches by progesterone analyses before the programmed mating improved the whelping rates. In the first experiment, the progesterone profiles of nulliparous and multiparous bitches were evaluated from Days 1 to 13 (onset of prooestrus = Day 0). The mean duration of the elevation in progesterone levels (> 2 ng/mL) after the onset of prooestrus tended to be approximately 1 day shorter in nulliparous bitches (7.7 days) than in multiparous bitches (8.5 days). In the second experiment, progesterone analyses in the bitches were carried out once on Days 4, 5 or 6. Bitches with progesterone levels of > 10 ng/mL were excluded from mating because it was unclear when the progesterone levels reached > 10 ng/mL considering the optimal date for mating. No significant differences were observed in the percentages of bitches excluded from mating and in the whelping rates of the mated bitches between the groups, irrespective of the day of progesterone analysis and the type of bitch. In conclusion, the initial elevation of progesterone levels was of shorter duration in nulliparous bitches. The selection of bitches by the measurement of progesterone levels once before mating was not effective for the programmed mating.

Background and Aims

Most people use the Internet in a functional way to achieve certain goals and needs. However, there is an increasing number of people who experience negative consequences like loss of control and distress based on an excessive use of the Internet and its specific online applications. Some approaches postulate similarities with behavioral addictions as well as substance dependencies. They differentiate between a generalized and a specific Internet addiction, such as the pathological use of social networking sites (SIA–SNS). Prior studies particularly identified the use of applications, personal characteristics, and psychopathological symptoms as significant predictors for the development and maintenance of this phenomenon. So far, it remains unclear how psychopathological symptoms like depression and social anxiety interact with individual expectancies of Internet use and capabilities of handling the Internet, summarized as Internet literacy.

Methods

The current study (N = 334) investigated the interaction of these components in a structural equation model.

Results

The results indicate that the effects of depression and social anxiety on SIA–SNS were mediated by Internet use expectancies and self-regulation.

Discussion

Thus, Internet use expectancies seem to be crucial for SIA–SNS, which is in line with prior models.

Conclusions

SNS use may be reinforced by experienced gratification and relief from negative feelings. Individual competences in handling the Internet may be preventive for the development of SIA–SNS.

The possibilities of lowering the immunoreactive properties of hydrolysed pea globulins were examined in the presented work. The hydrolytic process with Alcalase, pepsin and trypsin was carried out at temperatures of 37 °C and 50 °C for 180 min. The highest degree of hydrolysis was obtained during hydrolysis with Alcalase (DH 25% at temperature of 50 °C). The highest reduction of immunoreactive properties of legumin and vicilin was observed after hydrolysis with trypsin (DH 12%) at 50 °C by 98% and 97.4%, respectively. Generally, the immunoreactivity of vicilin was reduced to a smaller extent than immunoreactivity of legumin during the hydrolysis processes analysed.

Encephalitozoon intestinalis infection of sows is reported from a pig farm in Slovakia. Spores were detected by direct microscopic visualisation in the faeces of 25 out of 27 sows (92.6%). This finding was also supported serologically by the presence of specific anti-E. intestinalis antibodies and by a species-specific polymerase chain reaction (PCR). This is the first report on E. intestinalis infection of swine in Europe.
