In this study a simple and effective method was developed for the isolation of Saccharomyces strains from grapes. Aseptically collected grape samples were processed by enrichment in a nutritive basal medium supplemented with 10% (v/v) methanol followed by isolation of yeast strains. Sixteen of the 18 grape samples yielded Saccharomyces strain(s). More than 70% of the isolates belonged to the genus Saccharomyces. Based on phenotype and electrophoretic karyotyping, all strains of Saccharomyces were identified as S. cerevisiae. For several grape samples, varying physiological characters, the number of spores per asci, and the observed chromosome length polymorphisms provided evidence for diversity of S. cerevisiae strains obtained by this enrichment in methanol-containing broth. Results indicated that enrichment in methanol-containing broth is an effective alternative method to facilitate isolation of Saccharomyces strains from grapes. The enrichment method described in this work provides a simple and effective tool for isolation of Saccharomyces strains from grapes. The method may be applied in studying wine fermentation ecology, as well as for the isolation of potential starter strains from grapes.

Experiments were carried out on the sperm cryopreservation of artificially induced eels. The effects of several extenders and two cryoprotectants on the motility of spermatozoa were investigated. The highest post-thaw motility was observed with the combination of Tanaka's extender and DMSO as cryoprotectant. Further dilution after thawing resulted in complete loss of motility in samples frozen in presence of DMSO while sperm frozen with methanol as cryoprotectant retained its motility after further dilution.

Fenugreek (Trigonella foenum-graecum) of Fabacecae family is widely distributed throughout the world and used as an old medicinal plant and traditional food. The present study deals with the investigation of the anti-genotoxic potential of methanol (MTG) and ethanol (ETG) extracted leaf sap of fenugreek on Allium cepa root tip cells, which were treated with cadmium sulfate (CdSO4). Three types of treatments were applied. First, roots were treated with different concentrations of methanolic and ethanolic extracts (0.1%, 0.5% and 1%) separately for 3 h each, followed by CdSO4 treatment (at 250 ppm, for 3 h). Second, roots were first treated with CdSO4 followed by extracts treatment. Third, root tips were treated with CdSO4 with extracts treatments at the same time. For controls, roots with CdSO4 (250 ppm) and distilled water served as positive and negative control, respectively. The results showed that the methanol and ethanol extracts of fenugreek modulated the genotoxic and clastogenic aberrations, which were induced by CdSO4. The protection activity of MTG (1%) was 50% in the first treatment, 70% in the second treatment and 82% in the third treatment and 61%, 68% and 88% of ETG (1%), respectively. DNA rearrangements were also observed by revealing new RAPD bands in the total DNS samples isolated from Allium roots after treatmenst.

Xylopia aethiopica (XA) (Annonaceae) possesses great nutritional and medicinal values. This study was designed to investigate the effects of XA fruit methanol extract on oxidative stress in brain of rats exposed to whole body γ-radiation (5 Gy). Vitamin C (VC) served as standard antioxidant. Forty-four rats were divided into 4 groups of 11 rats each. One group served as control, two different groups were treated with XA and VC (250 mg/kg), 6 weeks before and 8 weeks after irradiation, and fourth group was only irradiated. Rats were sacrificed 1 and 8 weeks after irradiation. The antioxidant status, viz. Lipid peroxidation (LPO), superoxide dismutase (SOD), catalase (CAT), glutathione-s-transferase (GST) and glutathione (GSH) were estimated. Results indicate a significant increase ( p < 0.05) in levels of brain LPO after irradiation. LPO increased by 90% and 151%, after 1 and 8 weeks of irradiation, respectively. Irradiation caused significant ( p < 0.05) decreases in levels of GSH and GST by 61% and 43% after 1 week and, 75% and 73%, respectively, after 8 weeks of exposure. CAT and SOD levels were decreased by 62% and 68%, respectively, after 8 weeks of irradiation. Treatment with XA and VC ameliorated the radiation-induced decreases in antioxidant status of the animals. These suggest that XA could have beneficial effect by inhibiting oxidative damage in brain of exposed rats.
