Summary

In the present paper, a simple and reliable high-performance liquid chromatography-diode array detection (HPLC-DAD) method was developed both for quantitative determination and fingerprint analysis of Agrimonia pilosa Ledeb for quality control. Under the optimized HPLC conditions, seven bioactive compounds including rutin, quercetin-3-rhamnoside, luteoloside, tiliroside, apigenin, kaempferol, and agrimonolide were determined simultaneously. For fingerprint analysis, 11 common peaks were selected as the characteristic peaks to evaluate the similarities of 16 different samples collected from different origins in China. Besides, hierarchical cluster analysis (HCA) was also performed to evaluate the variation of the raw materials. This is the first report of using a simple method for quality control of A. pilosa Ledeb through multi-component determination and chromatographic fingerprint analysis to the best of our knowledge.

Summary

A simple and rapid method, using online ultraperformance liquid chromatography with photodiode array detection and electrospray ionization mass spectrometry (UPLC-PDA-eλ-ESI-MS/MS), was developed for the in-depth analysis of 50 batches Radix et Rhizoma Rhei. The analysis was performed on a UPLC BEH C18 column using a gradient elution system. Baseline separation could be achieved in less than 7.5 min. At the same time, on the basis of the 50 batches of samples collected from representative cultivated regions, a novel chromatographic fingerprint was devised by UPLC-PDA, in which 27 common peaks were detected and identified by the developed UPLC-MS/MS method step by step according to fragmentation mechanisms, MS/MS data, standards, and relevant literature. Many active components gave prominent [M - H] ions in the ESI mass spectra. These components include anthraquinones, sennosides, stilbenes, glucose gallates, naphthalenes, and catechins. Furthermore, based on the information of these Radix et Rhizoma Rhei components, and further combined with discriminant analysis, a novel discriminant analysis equation (DAE) was established for the quality control of Radix et Rhizoma Rhei for the first time.

In this paper, the wind energy resource in China’s oilfields is analyzed, the difference between China’s energy consumption structure and that of the world in average is analyzed as well, and the application prospect of wind heating technology in China’s oilfields is discussed as an example to reform China’s energy consumption structure. It shows that it is possible to use wind energy as an appropriate heat resource or supplementary heat source in some oilfields to supply heat energy for oil heating and living, more room to improve the energy resource utilization in various fields in China remain. It is a benefit to both oilfield and society to improve energy saving and environments.

Molecularly imprinted polymers (MIPs) were synthesized by imprinting a new template—S(-)-1,1′-binaphthalene-2,2′-diamine (S-DABN) and applied as chiral stationary phases for chiral separation of DABN racemates by high-performance liquid chromatography (HPLC). The influence of some key factors on the chiral recognition ability of MIPs, such as the type of functional monomers and porogen and the molar ratio of template to monomer, was systematically investigated. The chromatographic conditions, such as mobile phase composition, sample loading, and flow rate, were also measured. The chiral separation for DABN racemates under the optimum chromatographic conditions by using MIP chiral stationary phase (CSP) of P3, prepared with the S-DABN/MAA ratio = 1/4 and used acetonitrile (2 mL) and chloroform (4 mL) as porogen, showed the highest separation factor (2.14). Frontal analysis was used to evaluate affinity to the target molecule of MIPs. The binding sites (B t) of MIPs and dissociation constant (K d) were estimated as 4.56 μmol g−1 and 1.40 mmol L−1, respectively. In comparison with the previous studies, this approach had the advantages, such as the higher separation factor, easy preparation, and cost-effectiveness, it not only has the value for research but also has a potential in industrial application.

Growth and haemolytic activity of several pathogenic Vibrio species were compared in egg-fried-rice with different egg ratios. Egg-fried-rice preparations with rice-to-egg ratios of 4:1, 1:1, and 1:4 were inoculated with either Vibrio parahaemolyticus, V. cholerae, V. vulnificus, or V. alginolyticus and incubated for 24 h. Cell number, thermostable direct haemolysin (TDH) activity, and total haemolytic activity were determined. The cell number and total haemolytic activity increased in all Vibrio strains after 24 h, and these were most marked in egg-fried-rice with the highest egg content (1:4 (rice:egg) ratio; P<0.05). V. alginolyticus exhibited the maximal growth and V. parahaemolyticus the highest haemolytic activity, but only V. parahaemolyticus ATCC 33847, V. alginolyticus CAMT 21162, and V. alginolyticus HY 91101 showed TDH activity. Results suggest that lowering egg content in egg-fried-rice could reduce growth and virulence of Vibrio pathogens.

Elsholtzia densa Benth. var. densa (Lamiaceae) is a famous medicinal herb which has been widely used for treatment of colds, headaches, pharyngitis, fever, diarrhea, digestion disorder, rheumatic arthritis, nephritises, and nyctalopia in China. In this study, fraction of the ethyl alcohol extract of E. densa (aerial part) by different polarity solvents indicated that the ethyl acetate soluble fraction exhibited a potent 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity with the IC50 value of 148.2 μg/mL. Under the target guidance of DPPH experiment, isoquercitrin, trachelogenin, ethyl caffeate, and arctigenin were separated with purities 95.98%, 92.98%, 96.07%, and 88.83%, respectively, by a dual-mode high-speed counter-current chromatography (HSCCC) method using n-hexane–ethyl acetate–methanol–water (4.5:5:3:4, v/v/v/v) as the solvent system. In order to evaluate the scientific basis, antioxidant activity of four isolated compounds was assessed by the radical scavenging effect on DPPH radical; isoquercitrin and ethyl caffeate showed stronger antioxidant activities with IC50 values of 9.4 μg/mL and 9.2 μg/mL, respectively, while trachelogenin and arctigenin showed weak antioxidant activities with IC50 values of >500 μg/mL and 72.8 μg/mL, respectively. Results of the present study indicated that the combinative method using DPPH antioxidant assay and dual-mode HSCCC could be widely applied for rapid screening and isolating of antioxidants from complex traditional Chinese medicine extract.

Summary

To control the quality of Euonymus fortunei (Turcz.) Hand.-Mazz., a simple and reliable method of high-performance liquid chromatography (HPLC) coupled with photodiode array detector (PAD) was developed for both fingerprint analysis and quantitative determination. Four representative flavonoids, namely, kaempferol-3-O-β-D-glucopyranosyl-(1→4)-α-L-rhamnopyranosyl-7-O-β-D-glucopyranosyl-(1→4)-α-L-rhamnopyranoside (I), kaempferol-3,7-O-α-dirhamnopyranoside (II), apigenin-7-O-β-D-glucopyranoside (III), and kaempferol-3-(4″-O-acetyl)-O-α-L-rhamnopyranoside-7-O-α-L-r hamnopyranoside (IV) isolated from E. fortunei, were used as reference compounds and simultaneously determined by the validated HPLC method. The unique properties of the chromatographic fingerprint were validated by analyzing 11 batches of E. fortunei, E. japonicus, E. laxiflorus, E. myrianthus, and E. hamiltonianus samples. Our results revealed that the chromatographic fingerprint combined with similarity measurement could efficiently identify and distinguish E. fortunei from the other investigated Euonymus species.

Epimedium pubescens Maxim. and Epimedium koreanum Nakai. are two common and confused species of Herba Epimedii in Chinese Pharmacopoeia 2010 edition. Different species and growing conditions lead to chemical differences between the two species which may result in the improper clinical usage. In this work, a new method based on rapid-resolution liquid chromatography combined with time-of-flight mass spectrometry (RRLC/TOFMS) has been developed for identification and differentiation of major flavonoids in two kinds of Epimedium extract and rat plasma. The compounds were identified effectively based on the accurate extract masses and formulae acquired by RRLC/TOFMS. The fragmentation rules deduced by collision-induced dissociation (CID) were successfully implemented in distinguishing some of the isomers, further validating the results. By using the combined analytical techniques, a total of 40 major flavonoids in extracts of two kinds of Epimedium were identified within 30 min, including 31 common components and 9 characteristic components. After oral administration, three prototype compounds in rat plasma were detected by comparing the constituents measured in vitro with those in vivo, and five metabolites were identified by contrasting the fragmentation rules. The identification and structural elucidation of the chemical constituents provided essential data for further pharmacological and clinical studies on different species of Epimedium.

Abstract

Chinese onion (Allium chinense G. Don) is an edible vegetable as well as a traditional Chinese medicine. It is rich in steroidal saponins and possesses broad health benefits. In this study, the optimal extraction protocol of the total saponins from Chinese onion (ACS) was explored, and the content of the total steroidal saponins in ACS reached 56.62%. Network pharmacology was applied to predict the related signalling pathways and targets between the main phytochemicals in ACS and hypercholesterolaemia. Enrichment analysis showed that ACS might intervene hypercholesterolaemia through the PI3K-Akt signalling pathway. Meanwhile, cholesterol-lowering effects were verified by ACS intervention in high-cholesterol diet-induced hypercholesterolaemia in Kunming mice. Compared with the model group, the TC and LDL-C levels of mice were decreased and the HDL-C level increased significantly after administration of ACS at a dose of 200 mg kg−1 day−1. The body weight gain, liver index, and atherosclerosis index all decreased dramatically. ACS could significantly reduce the fat content in the liver and reduce the number of fat droplets from the haematoxylin and eosin (H&E) staining of mouse liver. The immunohistochemical staining indicated that ACS could up-regulate the expression of PI3K protein in the liver, thus playing an anti-hypercholesterolaemic role. This study indicated that ACS exhibited significant therapeutic and preventive effects on hypercholesterolaemia, and exerted anti-hypercholesterolaemia through the PI3K-Akt signalling pathway.

Abstract

Intermittent fasting (IF) is a dietary strategy that involves alternating periods of abstention from calorie consumption with periods of ad libitum food intake and has been shown to have beneficial effects in many ways. Recent studies have shown that IF attenuates neurodegeneration and improves cognitive decline, enhances functional recovery after stroke as well as attenuates the pathological and clinical features of epilepsy in animal models. Furthermore, IF induced several molecular and cellular adaptations in neurons that overall enhanced cellular stress resistance, synaptic plasticity, and neurogenesis. In this review, the beneficial effects of IF on central neurological disorders are discussed. The information summarised in this review can be used to help contextualise existing research and better guide the development of future IF interventions.
