Background and aims

Although smartphone use brings many benefits for adolescents, it is also associated with many serious health problems. This study examined the relationship between problematic smartphone use (PSU) and clinical health symptoms (e.g., body dysfunction) and the mediating effects of sleep quality on this relationship in adolescents.

Methods

Participants in this cross-sectional survey were 686 middle- and high-school students (girls = 55.7%, M age = 12.98 ± 1.38 years). Participants completed self-report measures of PSU, sleep quality, and physical symptoms. Correlation analyses and structural equation modeling between adolescents’ PSU and the variables of interest were conducted.

Results

This study indicated that there was a significant positive correlation between PSU and health symptoms. Furthermore, sleep quality mediated the relationship between PSU and health symptoms.

Discussion and conclusions

Findings suggest that to promote health and wellness in adolescents, individuals should be encouraged to place boundaries on smartphone use, especially at bedtime. Reducing adolescents’ exposure to smartphone use in this way may hold promise for improving the efficacy of PSU prevention efforts for adolescents.

Abstract

Background and aims

The working memory (WM) ability of internet addicts and the topology underlying the WM processing in internet addiction (IA) are poorly understood. In this study, we employed a graph theoretical framework to characterize the topological properties of the IA brain network in the source cortical space during WM task.

Methods

A sample of 24 subjects with IA and 23 matched healthy controls (HCs) performed visual 2-back task. Exact Low Resolution Electromagnetic Tomography was adopted to project the pre-processed EEG signals into source space. Subsequently, Lagged phase synchronization was calculated between all pairs of Brodmann areas, the graph theoretical approaches were then employed to estimate the brain topological properties of all participants during the WM task.

Results

We found better WM behavioral performance in IA subjects compared with the HCs. Moreover, compared to the HC group, more integrated and hierarchical brain network was revealed in the IA subjects in alpha band. And altered regional centrality was mainly resided in frontal and limbic lobes. In addition, significant relationships between the IA severity and the significant altered graph indices were found.

Conclusions

In conclusion, these findings provide evidence to support the notion that altered topological configuration may underline changed WM function observed in IA.

Patrinia scabiosaefolia Fisch. (PSF), a well-known traditional Chinese medicine, has been demonstrated to show therapeutic effects on inflammatory bowel disease. In this study, a rapid and sensitive method using ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC/Q-TOF-MS) was developed for identification of the major constituents in PSF. The separation analysis was performed on Waters Acquity UPLC system, and the accurate mass of molecules and their fragment ions were determined by Q-TOF-MS. Thirty-one constituents, including triterpenoids, iridoids, flavonoids, and organic acids were detected and tentatively deduced on the basis of their element compositions, tandem mass spectrometry (MS/MS) data, and relevant literatures. Twelve constituents were discovered for the first time in PSF. The results demonstrated that hederagenin-type and oleanolic acid-type saponins were the main constituents of PSF. Our work provides a certain foundation for further quantitation of major chemical constituents and in vivo pharmacokinetic studies of PSF. Moreover, the analytical approach developed herein has proven to be generally applicable for profiling the chemical constituents in traditional Chinese medicines (TCMs) and other complicated mixtures.

Abstract

A novel method was established for analysing trace four acidic phytohormones, namely, indole-3-acetic acid, 3-indolebutyric acid, abscisic acid, and 1-naphthylacetic acid, using magnetic ordered mesoporous carbon (MOMC). MOMC was facilely synthesised via self-assembly strategy with a direct carbonisation process. The properties of MOMC were characterised using various instruments. MOMC exhibited excellent adsorption capacity towards the analytes. Various critical parameters which may influence the enrichment efficiency were evaluated, including amount of MOMC, extraction conditions, and desorption conditions. An efficient method based on MOMC magnetic solid-phase extraction coupled with ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) was developed to analyse the trace four acidic phytohormones, with good correlation coefficients (R 2 = 0.9965–0.9998) and low limits of detection (0.13–9.7 ng L−1, S/N = 3). Trace acidic phytohormones in Agaricus bisporus and Hypsizygus marmoreus samples were determined with satisfactory recoveries (91.8–108%) and reproducibility (2.6–6.3%). The features indicated that MOMC provides an efficient platform for mushroom sampling; the developed method is convenient, promising, and sensitive for the detection of trace phytohormones in complicated mushroom samples.

Abstract

Background and aims

Mobile phone addiction (MPA) is emerging among adolescents, especially during the COVID-19 pandemic. Nevertheless, there is a dearth of knowledge regarding the correlation between MPA and suicide behaviors and its mechanism. The objective of the current study is exploring the direct effect of MPA on suicide behaviors and the indirect effect through poor sleep quality.

Methods

A total of 18,900 Chinese adolescents aged 12 to 18 were recruited via a multi-stage cluster sampling method.

Results

The prevalence of MPA and poor sleep quality was 26.2 and 23.1%, respectively. During the past year, 24.4% participants were involved in suicide behaviors. Specifically, suicide ideators, suicide planners, and suicide attempters were 10.7, 8.4, and 5.3%, respectively. Particularly, rural females had the highest prevalence of suicide behaviors, MPA, and poor sleep quality. Logistic regression analysis showed that MPA was significantly associated with suicide ideators (OR = 1.22, 95% CI: 1.09–1.37, p < 0.001) and planners (OR = 1.18, 95% CI: 1.04–1.34, p < 0.05), but not for suicide attempters (p > 0.05). Structural equation modelling demonstrated that MPA had direct effect on suicide behaviors (β = 0.145, 95% CI = 0.127–0.160), and poor sleep quality partially mediated the relationship (the mediating ratio was 46.7%). The mediating ratio of poor sleep quality was the highest in urban males.

Conclusions

MPA has both direct and indirect effects on suicide behaviors. For suicide prevention, limited mobile phone use and improvement sleep quality may be practical for adolescents. Additionally, more efforts of intervention could give priority to rural girls.

10-Methoxycamptothecin (MCPT) and 10-hydroxycamptothecin (HCPT) are the indole alkaloids isolated from a Chinese tree, Camptotheca acuminata, and have a wide spectrum of anticancer activity in vitro and in vivo mainly through inhibitory effects on topoisomerase I. HCPT is a major metabolite of MCPT in rats; the pharmacokinetic analysis and tissue distribution of MCPT and HCPT in rats have also been determined after i.v. injection of MCPT, but the excretion of MCPT and its metabolite HCPT has not been assessed up to now. In the present study, the excretion study of MCPT and its metabolite HCPT in rat bile, feces, and urine after i.v. administration of MCPT (5 mg kg−1) was performed by high-performance liquid chromatography (HPLC) method coupled with a fluorescence detector. The results showed that MCPT mainly biotransformed to HCPT and excreted in the form of HCPT and MCPT in bile, urine, and feces after i.v. administration of MCPT. It was excreted about 1.24 ± 0.07% as MCPT and 5.49 ± 0.40% as HCPT in bile within 6 h after i.v. administration. The cumulative excretions of MCPT and HCPT were mainly within 24 h after i.v. administration, which were 0.41 ± 0.10% and 7.66 ± 1.43% of the dosage in urine and about 0.16 ± 0.04% and 20.30 ± 3.35% of the dosage in feces. The total excretion of MCPT in urine, bile, and feces was 1.81 ± 0.09% in the form of original MCPT and 33.45 ± 1.57% detected as the metabolite HCPT in urine, bile, and feces, suggesting that MCPT might undergo other biotransformation.

Abstract

A rapid and sensitive High-Performance Liquid Chromatography-tandem Mass Spectrometry (HPLC/MS/MS) method for determining apremilast in beagle dog plasma and urine samples was developed and validated using clopidogrel as the internal standard (IS). Apremilast was extracted from the plasma and urine samples by liquid–liquid extraction using methyl tert-butyl ether. Chromatographic separation was performed using a C8 column with gradient elution and a mobile phase containing methanol and 0.1% formic acid. Quantification was achieved in multiple reaction monitoring (MRM) mode with a transition of m/z 461.3→178.2 for apremilast and m/z 322.2→184.1 for clopidogrel (IS). This method was validated regarding its specificity, linearity, precision, accuracy, and stability. The lower limit of quantification (LLOQ) for this method was 5 ng/mL, and the calibration curve was linear over 5–1,000 ng/mL. The intra- and inter-run coefficients of variance (CV) of aprelimast in plasma samples were less than 12.92% and 10.64%, respectively, while in urine samples, the CV were less than 11.84% and 10.20%, respectively. The samples were stable under the tested conditions. This method was successfully applied to a pharmacokinetic study in beagle dogs following oral administration of 10 mg of apremilast.

Abstract

Natural hemostatic compounds from Toddalia asiatica (Linn) Lam (T. asiatica) root bark had been investigated by a novel strategy, chemical fingerprint–pharmacokinetic–pharmacodynamic (CF–PK–PD) for the first time in this study. The extract sample of T. asiatica root bark was subdivided into petroleum ether (PE), ethyl acetate (EA) and n-butanol (n-B) sample by reagent extraction, EA sample showed significant hemostatic activity using prothrombin time (PT), activated partial thromboplastin time (APTT) and fibrinogen (FIB) as evaluation indexes from rat plasma of PK experiment in hemorrhagic rat model. CF analysis was adopted to assist us to discover six natural compounds from T. asiatica root bark in actual rat plasma after sample treatment by Ultra Performance Liquid Chromatography-Electrospray Ionization (UPLC-ESI) MS, there were only lomatin and 5-methoxy-8-hydroxy psoralen showing significant hemostatic effect (P < 0.05) mainly through endogenous coagulation pathway and fibrinolytic system. In PK–PD study, six compounds in EA sample exhibited relatively rapid absorption and slow elimination characteristics. The mean T max and t 1/2β of isopimpinellin and pimpinellin were 1.74 and 0.59 h, 5.31 and 6.89 h in rats. On the basis of Sigmoid–E max model, PK–PD related curves of FIB in hemorrhagic rat model after treatment of T. asiatica root bark were obtained. Predicted E max, EC 50 and k e0 of FIB under isopimpinellin were 4.87 mg/mL, 1.39 μg/mL and 0.81 1/h; predicted E max, EC 50 and k e0 of FIB under pimpinellin were 4.29 mg/mL, 2.47 μg/mL and 0.77 1/h. In conclusion, hemostatic compounds from T. asiatica root bark had been materialized, there were lomatin, isopimpinellin, pimpinellin and 5-methoxy-8-hydroxy psoralen at least as its main active substances through coagulation pathways and fibrinolytic system. CF–PK–PD method as a promising method was worthy of follow-up opening, application in pharmaceutical research.

High-performance liquid chromatography coupled with photodiode array detection and evaporative light scattering detection (HPLC—DAD—ELSD) was established to determine paeoniflorin and albiflorin simultaneously in Radix Paeoniae Rubra. The assay was performed on a Diamonsil C18 (4.6 mm × 250 mm, 5 μm) column by a gradient elution program with acetonitrile and aqueous formic acid (0.05% v/v) as mobile phase at a flow rate of 1.0 mL min−1. The detection wavelength of DAD was 230 nm, and the evaporator tube temperature of ELSD was set at 110 °C with the nebulizing gas flow rate of 3 L min−1. The temperature of column was kept at 30 °C. The linear ranges of paeoniflorin and albiflorin were within 0.050–1.510 mg mL−1 and 1.007–5.035 mg mL−1. The recoveries of paeoniflorin and albiflorin were 96.2–102.9% and 95.0–102.4%, respectively, while the relative standard deviation (RSD) of them was 0.2–2.5%. This method was quick, simple, accurate, and specific. It could be used for the quality control of Radix Paeoniae Rubra. The proposed approach was expected as a powerful tool for the quality control of Radix Paeoniae Rubra.

Background and aims

This large-scale study aimed to test (a) associations of problematic Internet use (PIU) and sleep disturbance with suicidal ideation and suicide attempts among Chinese adolescents and (b) whether sleep disturbance mediates the association between PIU and suicidal behavior.

Methods

Data were drawn from the 2017 National School-based Chinese Adolescents Health Survey. A total of 20,895 students’ questionnaires were qualified for analysis. The Young’s Internet Addiction Test was used to assess PIU, and level of sleep disturbance was measured by the Pittsburgh Sleep Quality Index. Multilevel logistic regression models and path models were utilized in analyses.

Results

Of the total sample, 2,864 (13.7%) reported having suicidal ideation, and 537 (2.6%) reported having suicide attempts. After adjusting for control variables and sleep disturbance, PIU was associated with an increased risk of suicidal ideation (AOR = 1.04, 95% CI = 1.03−1.04) and suicide attempts (AOR = 1.03, 95% CI = 1.02−1.04). Findings of the path models showed that the standardized indirect effects of PIU on suicidal ideation (standardized β estimate = 0.092, 95% CI = 0.082−0.102) and on suicide attempts (standardized β estimate = 0.082, 95% CI = 0.068−0.096) through sleep disturbance were significant. Conversely, sleep disturbance significantly mediated the association of suicidal behavior on PIU.

Discussion and conclusions

There may be a complex transactional association between PIU, sleep disturbance, and suicidal behavior. The estimates of the mediator role of sleep disturbance provide evidence for the current understanding of the mechanism of the association between PIU and suicidal behavior. Possible concomitant treatment services for PIU, sleep disturbance, and suicidal behavior were recommended.
