Summary
As a new form of traditional Chinese medicine (TCM), Zuojin pill extract is made containing Coptidis rhizoma (the rhizome of Coptis chinesis Franch. [Ranunculaceae]) and Euodiae fructus (the unripe fruit of Euodia rutaecarpa [Juss.] Benth. [Rutaceae]) at the ratio of 6:1 (w/w) and has been most widely used in TCM to treat gastro-intestinal disorders. However, the quality control is insufficient. For establishing an analysis method for the effective quality control of Zuojin pill extract, the active components of three protoberberine alkaloids (coptisine, palmatine, berberine, components from C. rhizoma) and two indolequinoline alkaloids (evodiamine, rutaecarpine, components from E. fructus) in Zuojin pill extract were determined simultaneously by high-performance liquid chromatography. Chromatographic separations were performed on an Inertsil ODS-3 column (4.6 mm × 250 mm, 5 μm). Elution was carried out at 25°C under isocratic conditions by using 25 mmol L−1 KH2PO4-25 mmol L−1 SDS-acetonitrile (1:1:2, v/v/v, adjusted pH to 3.0 using phosphoric acid) as a mobile phase with a flow rate of 0.5 mL min−1. Detection wavelength was set at 264 nm. Good linearity was obtained with correlation coefficient higher than 0.999 for all the analytes over the investigated concentration ranges. Three batches of Zuojin pill extract were successfully analyzed. The average percentages of coptisine, palmatine, berberine, evodiamine, and rutaecarpine in Zuojin pill extract were 5.35%, 3.92%, 16.64%, 0.03%, and 0.03%, respectively. Consequently, these protoberberine and indolequinoline alkaloids could be identified and determined easily by the established high-performance liquid chromatography (HPLC) method which can be used to evaluate and control the quality of Zuojin pill extract.
Epimedium pubescens Maxim. and Epimedium koreanum Nakai. are two common and confused species of Herba Epimedii in Chinese Pharmacopoeia 2010 edition. Different species and growing conditions lead to chemical differences between the two species which may result in the improper clinical usage. In this work, a new method based on rapid-resolution liquid chromatography combined with time-of-flight mass spectrometry (RRLC/TOFMS) has been developed for identification and differentiation of major flavonoids in two kinds of Epimedium extract and rat plasma. The compounds were identified effectively based on the accurate extract masses and formulae acquired by RRLC/TOFMS. The fragmentation rules deduced by collision-induced dissociation (CID) were successfully implemented in distinguishing some of the isomers, further validating the results. By using the combined analytical techniques, a total of 40 major flavonoids in extracts of two kinds of Epimedium were identified within 30 min, including 31 common components and 9 characteristic components. After oral administration, three prototype compounds in rat plasma were detected by comparing the constituents measured in vitro with those in vivo, and five metabolites were identified by contrasting the fragmentation rules. The identification and structural elucidation of the chemical constituents provided essential data for further pharmacological and clinical studies on different species of Epimedium.
Elsholtzia densa Benth. var. densa (Lamiaceae) is a famous medicinal herb which has been widely used for treatment of colds, headaches, pharyngitis, fever, diarrhea, digestion disorder, rheumatic arthritis, nephritises, and nyctalopia in China. In this study, fraction of the ethyl alcohol extract of E. densa (aerial part) by different polarity solvents indicated that the ethyl acetate soluble fraction exhibited a potent 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity with the IC50 value of 148.2 μg/mL. Under the target guidance of DPPH experiment, isoquercitrin, trachelogenin, ethyl caffeate, and arctigenin were separated with purities 95.98%, 92.98%, 96.07%, and 88.83%, respectively, by a dual-mode high-speed counter-current chromatography (HSCCC) method using n-hexane–ethyl acetate–methanol–water (4.5:5:3:4, v/v/v/v) as the solvent system. In order to evaluate the scientific basis, antioxidant activity of four isolated compounds was assessed by the radical scavenging effect on DPPH radical; isoquercitrin and ethyl caffeate showed stronger antioxidant activities with IC50 values of 9.4 μg/mL and 9.2 μg/mL, respectively, while trachelogenin and arctigenin showed weak antioxidant activities with IC50 values of >500 μg/mL and 72.8 μg/mL, respectively. Results of the present study indicated that the combinative method using DPPH antioxidant assay and dual-mode HSCCC could be widely applied for rapid screening and isolating of antioxidants from complex traditional Chinese medicine extract.
Abstract
Myricetin has been reported to have a wide variety of beneficial physiological functions. The present study investigated the antihyperlipidaemic activity of myricetin against hyperlipidaemia of high-fat diet-fed obese rats. The four-week antihyperlipidaemic activity was assayed by giving different doses of myricetin to hyperlipidaemic rats. Results showed that myricetin could reduce the harm caused by oxidative stress, decrease thiobarbituric acid reactive substances value, and decrease total cholesterol and triacylglycerol levels of hyperlipidaemic rats. Quantitative analysis of gene expression showed that myricetin's lipid-lowering activity can be activated by downregulating gene expression of fatty acid synthase (FAS) and peroxisome proliferator-activated receptor-γ (PPARγ) with upregulation of hormone-sensitive lipase (HSL) mRNA level. Thus, myricetin had significant health benefits and could be explored as a potentially promising dietary supplement for treating hyperlipidaemia.

A selective and sensitive liquid chromatography-electrospray ionization-mass spectrometry (HPLC-ESI-MS) method was developed and validated for analysis of xanthotoxol (1), xanthotoxin (2), isoimpinellin (3), bergapten (4), oxypeucedanin (5), imperatorin (6), cnidilin (7), and isoimperatorin (8) in rat bile and urine using pimpinellin as an internal standard (IS). An Agilent 1200 liquid chromatography system (Agilent Technologies, USA) equipped with a quaternary pump, an autosampler, and a column compartment was used for all analyses. Chromatographic separations were performed on a Sapphire C18 column (150 mm × 4.6 mm, 5 μm), and the column temperature was maintained at 30°C; the sample injection volume was 10 μL. The specificity, linearity, accuracy, precision, recovery, matrix effect, and several stabilities were validated for all analytes in the rat bile and urine samples. The method was successfully applied in monitoring the concentrations of eight coumarins in rat bile and urine after a single oral administration of Radix Angelicae Dahuricae extract with a dosage of 8.0 mL/kg. In the bile samples, the eight coumarins excreted completely in twenty-four hours. The average percentages of coumarins (1–8) excreted were 0.045%, 0.019%, 0.177%, 0.105%, 0.337%, 0.023%, 0.024%, 0.021%. In the urine samples, the eight coumarins excreted completely in seventy-two hours. The average percentages of coumarins (1–8) excreted were 1.78%, 0.095%, 0.130%, 0.292%, 0.082%, 0.008%, 0.005%, 0.004%. The method is robust and specific and it can successfully complete the requirements of the excretion study of the eight coumarins in Radix Angelicae Dahuricae.
Abstract
The work was carried out to explore whether the anti-inflammatory effect of n-3 unsaturated fatty acids on patients with rheumatoid arthritis is related to the levels of inflammatory markers CRP and ESR. Studies on the treatment of rheumatoid arthritis with n-3 unsaturated fatty acid diet therapy and the outcome index containing CRP and/or ESR were included, and studies on the subjects suffering from other diseases affecting the outcome index were excluded. PubMed, Web of Science, EMBASE and Google Scholar were systematically searched, and all studies published from the establishment of the library to 2024 were collected. The Cochrane Bias Risk Assessment tool was used to evaluate the quality of the included studies. Data analysis was performed using Stata 16.0 software. Seven studies were included in this analysis. A total of 399 subjects were studied to explore the effect of an n-3 unsaturated fatty acid diet on rheumatoid arthritis. The results showed that there was no significant difference in CRP (Hedges's g = 0.06, 95% CI: −0.48–0.37, P = 0.79) and ESR (Hedges's g = −0.14, 95% CI: −0.61–0.33, P = 0.55) between the intervention and control groups. The results of this study showed that the anti-inflammatory effect of unsaturated fatty acids on rheumatoid arthritis was not correlated with CRP and ESR levels. Due to the small number of included studies, more high-quality studies are still needed to confirm this.
