Abstract

This work used a carrageenan-based thrombosis model to determine the preventative effects of Lactobacillus plantarum YS1 (LPYS1) on thrombus. In thrombotic mice, LPYS1 improved the activated partial thromboplastin time (APTT), while decreasing the thrombin time (TT), prothrombin time (PT), and fibrinogen (FIB) content. In thrombotic mouse serum, LPYS1 decreased the levels of malondialdehyde (MDA), tumour necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), nuclear factor kappa-B (NF-κB), and interleukin-1 beta (IL-1β), while also increasing the activities of superoxide dismutase (SOD) and catalase (CAT). Moreover, LPYS1 upregulated the mRNA expression levels of copper/zinc-SOD (Cu/Zn-SOD), manganese-SOD (Mn-SOD), and CAT in the colon tissues of thrombotic mice, while downregulating those of NF-κB p65, IL-6, TNF-α, and interferon-gamma (IFN-γ) mRNA. In tail vein vascular tissues, LPYS1 suppressed the mRNA expression levels of NF-κB p65, intercellular cell adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. The abundances of both beneficial and pathogenic bacteria were altered by LPYS1. These findings show that LPYS1 has the capacity to protect mice from thrombosis, while also revealing some of the underlying mechanisms of this effect.

Abstract

Chinese onion (Allium chinense G. Don) is an edible vegetable as well as a traditional Chinese medicine. It is rich in steroidal saponins and possesses broad health benefits. In this study, the optimal extraction protocol of the total saponins from Chinese onion (ACS) was explored, and the content of the total steroidal saponins in ACS reached 56.62%. Network pharmacology was applied to predict the related signalling pathways and targets between the main phytochemicals in ACS and hypercholesterolaemia. Enrichment analysis showed that ACS might intervene hypercholesterolaemia through the PI3K-Akt signalling pathway. Meanwhile, cholesterol-lowering effects were verified by ACS intervention in high-cholesterol diet-induced hypercholesterolaemia in Kunming mice. Compared with the model group, the TC and LDL-C levels of mice were decreased and the HDL-C level increased significantly after administration of ACS at a dose of 200 mg kg−1 day−1. The body weight gain, liver index, and atherosclerosis index all decreased dramatically. ACS could significantly reduce the fat content in the liver and reduce the number of fat droplets from the haematoxylin and eosin (H&E) staining of mouse liver. The immunohistochemical staining indicated that ACS could up-regulate the expression of PI3K protein in the liver, thus playing an anti-hypercholesterolaemic role. This study indicated that ACS exhibited significant therapeutic and preventive effects on hypercholesterolaemia, and exerted anti-hypercholesterolaemia through the PI3K-Akt signalling pathway.

Abstract

The work was carried out to explore whether the anti-inflammatory effect of n-3 unsaturated fatty acids on patients with rheumatoid arthritis is related to the levels of inflammatory markers CRP and ESR. Studies on the treatment of rheumatoid arthritis with n-3 unsaturated fatty acid diet therapy and the outcome index containing CRP and/or ESR were included, and studies on the subjects suffering from other diseases affecting the outcome index were excluded. PubMed, Web of Science, EMBASE and Google Scholar were systematically searched, and all studies published from the establishment of the library to 2024 were collected. The Cochrane Bias Risk Assessment tool was used to evaluate the quality of the included studies. Data analysis was performed using Stata 16.0 software. Seven studies were included in this analysis. A total of 399 subjects were studied to explore the effect of an n-3 unsaturated fatty acid diet on rheumatoid arthritis. The results showed that there was no significant difference in CRP (Hedges's g = 0.06, 95% CI: −0.48–0.37, P = 0.79) and ESR (Hedges's g = −0.14, 95% CI: −0.61–0.33, P = 0.55) between the intervention and control groups. The results of this study showed that the anti-inflammatory effect of unsaturated fatty acids on rheumatoid arthritis was not correlated with CRP and ESR levels. Due to the small number of included studies, more high-quality studies are still needed to confirm this.
