Aegiolops kotschyi cytoplasmic male sterile system often results in part of haploid plants in wheat (Triticum aestivum L.). To elucidate the origin of haploid, 235 wheat microsatellite (SSR) primers were randomly selected and screened for polymorphism between haploid (2n = 3x = 21 ABD) and its parents, male-sterile line YM21 (2n = 6x = 42 AABBDD) and male fertile restorer YM2 (2n = 6x = 42 AABBDD). About 200 SSR markers yielded clear bands from denatured PAGE, of which 180 markers have identifiable amplification patterns, and 20 markers (around 8%) resulted in different amplification products between the haploid and the restorer, YM2. There were no SSR markers that were found to be distinguishable between the haploid and the male sterile line YM21. In addition, different distribution of HMW-GS between endosperm and seedlings from the same seeds further confirmed that the haploid genomes were inherited from the maternal parent. After haploidization, 1.7% and 0.91% of total sites were up- and down-regulated exceeding twofold in the shoot and the root of haploid, respectively, and most of the differentially expressed loci were up/down-regulated about twofold. Out of the sensitive loci in haploid, 94 loci in the shoot, 72 loci in the root can be classified into three functional subdivisions: biological process, cellular component and molecular function, respectively.

The migration of 99Tc in unsaturated Chinese loess was investigated in-situ with a tracer method. Quartz containing 3H (HTO) and 99Tc (99TcO4-) was introduced into the bottom of an experimental pit which was then backfilled at the field test site. Then core soil samples were taken and cut vertically into 1 cm long slices. The slice samples were analyzed by liquid scintillation techniques in the laboratory. The results indicate that the migration pattern of 99Tc was quite similar to that of 3H and the vertical diffusion coefficients of 99Tc and 3H were calculated as (4.7±0.4).10-2 cm2/d and (7.8±0.4).10-2 cm2/d, respectively.

Elsholtzia densa Benth. var. densa (Lamiaceae) is a famous medicinal herb which has been widely used for treatment of colds, headaches, pharyngitis, fever, diarrhea, digestion disorder, rheumatic arthritis, nephritises, and nyctalopia in China. In this study, fraction of the ethyl alcohol extract of E. densa (aerial part) by different polarity solvents indicated that the ethyl acetate soluble fraction exhibited a potent 1,1-diphenyl-2-picryhydrazyl (DPPH) radical scavenging activity with the IC50 value of 148.2 μg/mL. Under the target guidance of DPPH experiment, isoquercitrin, trachelogenin, ethyl caffeate, and arctigenin were separated with purities 95.98%, 92.98%, 96.07%, and 88.83%, respectively, by a dual-mode high-speed counter-current chromatography (HSCCC) method using n-hexane–ethyl acetate–methanol–water (4.5:5:3:4, v/v/v/v) as the solvent system. In order to evaluate the scientific basis, antioxidant activity of four isolated compounds was assessed by the radical scavenging effect on DPPH radical; isoquercitrin and ethyl caffeate showed stronger antioxidant activities with IC50 values of 9.4 μg/mL and 9.2 μg/mL, respectively, while trachelogenin and arctigenin showed weak antioxidant activities with IC50 values of >500 μg/mL and 72.8 μg/mL, respectively. Results of the present study indicated that the combinative method using DPPH antioxidant assay and dual-mode HSCCC could be widely applied for rapid screening and isolating of antioxidants from complex traditional Chinese medicine extract.

A recombinant inbred line (RIL) population with 302 lines derived from a cross of Weimai 8 × Luohan 2 was used to identify the quantitative trait loci (QTL) for plant height (PH) in wheat (Triticum aestivum L.). Possible genetic relationships between PH and PH components (PHC), including spike length (SL) and internode length from the first to the fourth node counted from the top, abbreviated as FIITL, SITL, TITL and FOITL, respectively, were evaluated at the QTL level. A QTL for PH was mapped using data on PH and on PH conditioned by PHC using the IciMapping V3.0 software. Conditional QTL mapping proved that, at the QTL level, SL contributed the least to PH, followed by FIITL and FOITL, while TITL had the strongest influence on PH, followed by SITL. These results indicate that the conditional QTL mapping method can be used to evaluate possible genetic relationships between PH and PHC, and that it can efficiently and precisely reveal counteracting QTL, which will enhance our understanding of the genetic basis of PH in wheat.

The effects of fullerenes, including fellerene soot (FS), extracted fullerene soot (EFS) and pure C60 on the thermal decomposition of ammonium perchlorate (AP) compared with traditional carbon black (CB) catalyst has been studied
by employing thermogravimetry (TG), differential thermal analysis (DTA), infrared spectroscopy (IR) and ignition temperature
experiments. The results showed that the addition of CB and FS to AP reduced the activation energy as well as the temperature
at maximum decomposition rate, but that of EFS and pure C60 had little effect on the thermal decomposition of AP, and among all catalysts, FS was the best one.

Stripe rust, caused by Puccinia striiformis Westend. f. sp. tritici Eriks., is a major disease that causes substantial losses to wheat production worldwide. The utilization of effective resistance genes in wheat cultivars is the preferred control of the disease. To study the inheritance of all-stage resistance in spring wheat cultivars Louise, WA008016, Express, Solano, Alturas and Zak from the Pacific Northwest (PNW) of the United States, the six cultivars were crossed with the Chinese susceptible variety Taichung 29. Single-spore isolates of CYR32 and CYR33, the predominant Chinese races of P. striiformis f. sp. tritici, were used to evaluate F1, F2 and BC1 generations for stripe rust resistance under controlled greenhouse conditions. Genetic analysis determined that Louise had one dominant resistance gene to CYR32, temporarily designated as YrLou. WA008016 had two dominant and one recessive resistance genes to CYR32, temporarily designated as YrWA1, YrWA2 and YrWA3, respectively. Express had a single recessive gene that conferred resistance to CYR32, temporarily designated as YrExp3. The two independent dominant genes in Solano conferring resistance to CYR32 were temporarily designated as YrSol1 and YrSol2. Alturas had two recessive genes for resistance to CYR32, temporarily designated as YrAlt1 and YrAlt2. Zak has one dominant gene for resistance to CYR33, temporarily designated as YrZak1. These six cultivars can be important resistance sources in Chinese wheat stripe rust resistance breeding.

A method was developed for the preparative separation of two alkaloids from the crude extract of the radix of Rauvolfia verticillata (Lour.) Baill. in a single run. The two-phase solvent system composed of petroleum ether–ethyl acetate–methanol–water (5:5:2:8, v/v), where triethylamine (40 mmol/L) was added to the upper organic phase as the stationary phase and hydrochloric acid (10 mmol/L) was added to the lower aqueous phase as the mobile phase, was selected for this separation by pH-zone-refining counter-current chromatography (PZRCCC). For the preparative separation, the apparatus was rotated at a speed 850 rpm, while the mobile phase was pumped into the column at 2 mL/min. As a result, 112 mg of reserpine and 21 mg of yohimbine were obtained from 3 g of crude extract in a single run. The analysis of the isolated compounds was determined by high-performance liquid chromatography (HPLC) at 230 nm with purities of over 91.0%, and the chemical identification was carried out by the data of electrospray ionization–mass spectrometry (ESI–MS) and nuclear magnetic resonance (NMR) spectroscopy. The technique introduced in this paper is an efficient method for preparative separation of reserpine and yohimbine from devil pepper radix. It will be beneficial to utilize medicinal materials and also useful for the separation, purification, and pharmacological study of Chinese herbal ingredients.

Thinopyrum intermedium, which has many useful traits, is valuable for wheat breeding. A new wheat-Thinopyrum addition line, SN100109, was developed from the progeny of common wheat cultivar Yannong 15 and Th. intermedium. It was resistant to most races of Blumeria graminis f. sp tritici (Bgt), which caused powdery mildew in wheat, and its reactions were different from the reactions of gene Pm40 and Pm43. Genomic in situ hybridization (GISH) and molecular marker analysis were used to identify the genomic composition of SN100109. GISH results showed that SN100109 was a wheat-Th. intermedium disomic addition line containing one pair of J chromosomes, and the resistance gene was located on the alien additional chromosomes of SN100109. And four molecular markers BE425942, BF482714, Xgdm93 and BV679214 which were assigned to homologous group 2, were specific molecular markers of the additional chromosomes. All the results indicated that SN100109 contained one pair of 2J chromosomes. SN100109 can be used as a novel germplasm source for introducing powdery mildew resistance genes to wheat in breeding programs.

To develop thermal stable flavor, two glycosidic bound flavor precursors, geranyl-tetraacetyl-β-D-glucopyranoside (GLY-A) and geranyl-β-D-glucopyranoside (GLY-B) were synthesized by the modified Koenigs–Knorr reaction. The thermal decomposition process and pyrolysis products of the two glycosides were extensively investigated by thermogravimetry (TG), differential scanning calorimeter (DSC) and on-line pyrolysis-gas chromatography mass spectroscopy (Py-GC-MS). TG showed the Tp of GLY-A and GLY-B were 254.6 and 275.7°C. The Tpeak of GLY-A and GLY-B measured by DSC were 254.8 and 262.1°C respectively.
Py-GC-MS was used for the simply qualitative analysis of the pyrolysis products at 300 and 400°C. The results indicated that: 1) A large amount of geraniol and few by-products were produced at 300°C, the by-products were significantly increased at 400°C; 2) The characteristic pyrolysis product was geraniol; 3) The primary decomposition reaction was the cleavage of O-glycosidic bound of the two glycosides flavor precursors. The study on the thermal behavior and pyrolysis products of the two glycosides showed that this kind of flavor precursors could be used for providing the foodstuff with specific flavor during heating process.

High-yield common buckwheat ‘cv. Fengtian 1’ (FT1) and tartary buckwheat ‘cv. Jingqiao 2’ (JQ2) were selected to investigate the characteristics of the grain-filling process and starch accumulation of high-yield buckwheat. FT1 had an average yield that was 43.0% higher than that of the control ‘cv. Tongliaobendixiaoli’ (TLBDXL) in two growing seasons, while JQ2 had an average yield that was 27.3% higher than that of the control ‘cv. Chuanqiao 2’ (CQ2). The Richards equation was utilized to evaluate the grain-filling process of buckwheat. Both FT1 and JQ2 showed higher values of initial growth power and final grain weight and longer linear increase phase, compared with respective control. These values suggest that the higher initial increasing rate and the longer active growth period during grain filling play important roles to increase buckwheat yield. Similar patterns of starch, amylose and amylopectin accumulation were detected in common buckwheat, leading to similar concentration of each constituent at maturity in FT1 and TLBDXL. Tartary buckwheat showed an increasing accumulation pattern of amylose in developing seeds, which differed from that of starch and amylopectin. This pattern led to a significant difference of the concentrations of amylose and amylopectin at maturity between JQ2 and CQ2, the mechanisms of which remained unclear. Nevertheless, both FT1 and JQ2 showed increased starch, amylose, and amylopectin accumulation during the physiological maturity of grains. The results suggest that prolonging the active grain-filling period to increase carbohydrate partitioning from source to seed sink can be an effective strategy to improve buckwheat yield.
