Effects of hydrocolloids (arabic gum, guar gum, and xanthan gum) on the physicochemical and rheological properties of whole-barley fortified cracker flour were determined using solvent retention capacity, alveograph, and Mixolab profiles. Results showed that the water absorption of whole-barley fortified cracker flour was reduced by the additional arabic gum. Besides, arabic gum was more effective in reducing the resistance to inflation and improving the extensibility of whole-barley fortified dough. Mixolab parameters indicated that the weakening of gluten proteins and the rate of starch retrogradation in whole-barley fortified cracker dough were reduced by the presence of arabic gum. Guar gum and xanthan gum promoted the rate of protein breakdown, but slowed down the starch gelatinization and retrogradation rate during the Mixolab heating-cooling cycle. In conclusion, involved arabic gum rather than guar gum or xanthan gum is benefit to improve the baking quality of wholebarley fortified saltine crackers.

The aerial surface of land plants is protected by a cuticle against abiotic and biotic stresses. A better understanding of the determinants of cuticle formation and function has the potential to contribute to the breeding of more drought tolerant and disease resistant crop varieties. Two doubled haploid (DH) mapping populations, Steptoe × Morex populations and OWB-dominant × recessive populations were exploited to genetically position homologs of a set of known cuticle-associated genes. These genes were also placed on a consensus map, BinMap2005, which includes 27 eceriferum (cer) loci. Of the 49 known cuticle-associated genes, 21 identified a homolog in barley, and of these, 14 were mapped. There was a complete linkage between HvCER6 and cer-zg, suggesting the possibility that HvCER6 is the candidate gene of CER-ZG. Positioning known cuticle-associated genes on a consensus map containing cuticle mutant loci may guide the selection of candidate genes for cuticle mutants, and thus facilitate the isolation of cuticle-associated genes in barley.

Abstract  

A novel solid complex, formulated as Ho(PDC)3 (o-phen), has been obtained from the reaction of hydrate holmium chloride, ammonium pyrrolidinedithiocarbamate (APDC) and 1,10-phenanthroline (o-phenH2O) in absolute ethanol, which was characterized by elemental analysis, TG-DTG and IR spectrum. The enthalpy change of the reaction of complex formation from a solution of the reagents, Δr H m θ (sol), and the molar heat capacity of the complex, c m, were determined as being –19.1610.051 kJ mol–1 and 79.2641.218 J mol–1 K–1 at 298.15 K by using an RD-496 III heat conduction microcalorimeter. The enthalpy change of complex formation from the reaction of the reagents in the solid phase, Δr H m θ(s), was calculated as being (23.9810.339) kJ mol–1 on the basis of an appropriate thermochemical cycle and other auxiliary thermodynamic data. The thermodynamics of reaction of formation of the complex was investigated by the reaction in solution at the temperature range of 292.15–301.15 K. The constant-volume combustion energy of the complex, Δc U, was determined as being –16788.467.74 kJ mol–1 by an RBC-II type rotating-bomb calorimeter at 298.15 K. Its standard enthalpy of combustion, Δc H m θ, and standard enthalpy of formation, Δf H m θ, were calculated to be –16803.957.74 and –1115.428.94 kJ mol–1, respectively.

Summary  

The removal of heavy metal ion Co(II) from aqueous solution was studied usingg-Al2O3 by batch technique. The experiments were performed at T=20±2 °C, in 0.01M NaNO3 solutions and under aerobic conditions. The effect of pH, ionic strength, fulvic acid (FA) and alumina amount on the sorption of Co(II) on alumina were also investigated. The pH affected the sorption of Co(II) significantly as compared with the effect of FA and ionic strength. The results indicated that strong chemical bonds are formed between Co(II) and functional groups of the bare or FA coated alumina, and a precipitation of Co(II) takes place on the alumina surface, induced by a transition from the adsorption to surface. The addition sequences of Co/FA on Co(II) sorption were also studied: the sorption of Co(II) in the ternary system was found independent of addition sequences.

Abstract  

On the basis of the theory of thermokinetics proposed in the literature, a novel thermokinetic method for determination of the reaction rate, the characteristic parameter method, is proposed in this paper. Mathematical models were established to determine the kinetic parameters and rate constants. In order to test the validity of this method, the saponifications of ethyl benzoate, ethyl acetate and ethyl propionate, and the formation of hexamethylenetetramine were studied with this method. The rate constants calculated with this method are in agreement with those in the literature, and the characteristic parameter method is therefore believed to be correct.In the light of the characteristic parameter method, we have developed further two thermo-kinetic methods, the thermoanalytical single and multi-curve methods, which are convenient for simultaneous determination of the reaction order and the rate constant. The reaction orders and rate constants of the saponifications of ethyl acetate and ethyl butyrate and the ring-opening reaction of epichlorohydrin with hydrobromic acid were determined with these methods, and their validity was verified by the experimental results.

Summary

A simple and rapid HPLC method using a photodiode array (PDA) detector for the analysis of 3-hydroxycarboplatin and its related complex has been established for the first time. Separation of 3-hydroxycarboplatin and 3-hydroxy-1,1-cyclobutanedicarboxylic acid (3-HO-cbdca) was carried out on a Phenomenex ODS3 column using an aqueous solution containing 50 mM ammonium acetate and 5 mM sodium 1-octanesulfonate as the mobile phase. The flow rate was 0.8 mL min−1, the column temperature was 40°C, and the detection wavelength was 230 nm for 3-hydroxycarboplatin and 220 nm for 3-HO-cbdca. Different analytical performance parameters such as precision, accuracy, linearity, stability of the solution, specificity, limit of detection (LOD), limit of quantification (LOQ), and system suitability were determined using the Empower 2 software. The calibration curve of standard 3-hydroxycarboplatin showed good linearity (r = 0.9995) within the range 0.5–1.4 mg mL−1. The method was accurate and precise, with an average accuracy of 100.4% (RSD = 1.53%, n = 9), and the results of the system suitability test showed symmetrical peaks, good resolution (R s), and repeatability. It can be applied to the quality control of 3-hydroxycarboplatin.

Effects of pH, ionic strength and fulvic acid on sorption of radiocobalt on montmorillonite and its Al-pillared and cross-linked samples were studied using batch technique. The results indicate that the sorption of cobalt is strongly dependent on pH values and independent of ionic strength. Fulvic acid enhances the sorption of cobalt slightly at low pH, but has no influence at high pH values. Surface complexation is considered the main mechanism of cobalt sorption to montmorillonite. The sequences of FA/Co2+ additions to the system did not affect cobalt sorption.

To provide a convenient and facile method to evaluate the radiochemical purity (RCP) of 99mTc-TRODAT-1 in quality control of routine clinical application, a simplified method of single-strip thin layer chromatography (TLC) was developed and validated by high performance liquid chromatography (HPLC). The RCP data of TLC correlated well with HPLC.

Two new y-type HMW-GSs in Ae. tauschii , 1Dy12.1* t and 1Dy12.2 t with the mobility order of 1Dy12.2 t > 1Dy12.1* t > 1Dy12.1 t >1Dy12, were identified by both SDS-PAGE and MALDI-TOF-MS. Molecular cloning and sequencing showed that the genes encoding subunits 1Dy12.1* t and 1Dy12.2 t had identical nucleotide acid sequences with 1,947 bp encoding a mature protein of 627 residues. Their deduced molecular weights were 67,347.6 Da, satisfactorily corresponding to that of 1Dy12.2 t subunit determined by MALDI-TOF-MS (67,015.7 Da), but was significantly smaller than that of the the 1Dy12.1* t subunit (68,577.1 Da). Both subunits showed high similarities to 1Dy10, suggesting that they could have a positive effect on bread-making quality. Interestingly, the expressed protein of the cloned ORF from accessions TD87 and TD130 in E. coli co-migrated with subunit 1Dy12.2 t , but moved slightly faster than 1Dy12.1* t on SDS-PAGE. The expressed protein in transgenic tobacco seeds, however, had the same mobility as the 1Dy12.1* t subunit, as confirmed by both SDS-PAGE and Western blotting. Although direct evidence of phosphoprotein could not be obtained by specific staining method, certain types of post-translational modifications (PTMs) of the 1Dy12.1* t subunit could not be excluded. We believe PTMs might be responsible for the molecular weight difference between the subunits 1Dy12.1* t and 1Dy12.2 t .
