Gliadin is a main component of gluten proteins that affect functional properties of bread making and contributes to the viscous nature of doughs. In this study, thirteen novel ω-gliadin genes were identified in several Triticum species, which encode the ARH-, ATDand ATN-type proteins. Two novel types of ω-gliadins: ATD- and ATN- have not yet been reported. The lengths of 13 sequences were ranged from 927 to 1269 bp and the deduced mature proteins were varied from 309 to 414 residues. All 13 genes were pseudogenes because of the presence of internal stop codons. The primary structure of these ω-gliadin genes included a signal peptide, a conserved N-terminal domain, a repetitive domain and a conserved C-terminus. In this paper, we first characterize ω-gliadin genes from T. timopheevi ssp. timopheevi and T. timopheevi ssp. araraticum. The ω-gliadin gene variation and the evolutionary relationship of ω-gliadin family genes were also discussed.

Campylobacter jejuni’s flagellar locomotion is controlled by eleven chemoreceptors. Assessment of the distribution of the relevant chemoreceptor genes in the C. jejuni genomes deposited in the National Center for Biotechnology Information (NCBI) database led to the identification of two previously unknown tlp genes and a tlp5 pseudogene. These two chemoreceptor genes share the same locus in the C. jejuni genome with tlp4 and tlp11, but the gene region encoding the periplasmic ligand binding domain differs significantly from other chemoreceptor genes. Hence, they were named tlp12 and tlp13.

Consequently, it was of interest to study their distribution in C. jejuni subpopulations of different clonality, and their cooccurrence with the eleven previously reported chemoreceptor genes. Therefore, the presence of all tlp genes was detected by polymerase chain reaction (PCR) in 292 multilocus sequence typing (MLST)-typed C. jejuni isolates from different hosts.

The findings show interesting trends: Tlp4, tlp11, tlp12, and tlp13 appeared to be mutually exclusive and cooccur in a minor subset of isolates. Tlp4 was found to be present in only 33.56% of all tested isolates and was significantly less often detected in turkey isolates. Tlp11 was tested positive in only 17.8% of the isolates, while tlp12 was detected in 29.5% of all isolates, and tlp13 was found to be present in 38.7%.

Recently, four novel genes named Pinb-2, with 57–60% sequence similarity with wild-type allele Pinb-D1a coding for grain-hardness related puroindoline B have been shown to occur on homoeologous group 7 chromosomes in bread wheat (Triticum aestivum). In the present report, T. monococcum ssp. monococcum (Am genome) revealed a Pinb-2 gene with a poly-G tract and an in-frame TAG stop codon at the 5′ terminus of the coding DNA sequence. The stop codon was observed in 53 accessions of different geographic origins, suggesting that Pinb-2 in ‘monococcum’ wheat is unlikely to be expressed. By contrast, the coding DNA sequence of Pinb-2 in T. urartu (Au genome) was found to be 99% identical to its counterpart on chromosome 7AL in bread and durum (T. turgidum ssp. durum) wheat. Moreover, a sequence very similar to “urartu” Pinb-2 was found in tetraploid wheat T. timopheevii and hexaploid wheat T. zhukovskyi. This latter species exhibited an additional Pinb-2 pseudogene inherited from T. monococcum. The results are discussed in relation to the lineage of T. zhukovskyi and the potential role of Pin-b2 on kernel texture.
