A simple thin-layer chromatographic method suitable for monitoring microbiological fermentation processes is reported. In addition to compounds related to the target compound the method enables acquisition of data on the formation and degradation of a series of carbohydrates from starch to monosaccharides. The target compounds and the carbohydrates can both be analyzed by use of a single method using one layer only. Both sets of data are useful for optimization of different biosyntheses or bioconversions. The method involves one, effective, thin-layer chromatographic separation, subsequent charring with sulfuric acid solution as detection reagent, heating, and evaluation by scanning the colored spots.

A mouflon showing severe weight loss and cachexia was examined. The animal had horns and a male-like body frame but the preputial fur was missing. The scrotum was completely absent. Both testicles were located next to the inguinal canal under the skin, and appeared only slightly smaller than normal. The prepuce was located in a perianal position under the anus and it resembled a vulva. The penis was underdeveloped and curled up inside the prepuce like an enlarged clitoris. The bulbourethral region and the urinary bladder looked normal. The orifice of the urethra was located in its normal position, but the glans penis appeared deformed. Microscopic examination of the testicles revealed mild degeneration of the seminiferous tubules and a marked proliferation of the interstitial connective tissue with Leydig cells still present. There were no marked changes in the bulbar part of the penis but the apical part exhibited marked interstitial fibrosis. This is the first description of hypospadias and cryptorchidism in mouflon.

TLC methods have been developed for monitoring microbial enantioselective reduction and regioselective oxidation. The work discussed in this paper focused on preparation of aromatic chiral alcohols by bioreduction of their respective acetone derivatives by biocatalysis using yeast strains in medium containing added resin. A simple method and suitable device were developed for sampling from such heterogeneous fermentation media. Methods and chromatograms are presented for quantitation by TLC and for enantiomer ratio determination by HPLC. With regard to oxidation procedures, a TLC method was established for monitoring compactin hydroxylation during fermentation processes — as a representative example. Improvement of the method and use of HPTLC plates resulted in high resolution and separation capacity equal to that of the HPLC method. By means of the method by-products usually formed as minor components in the biotransformation could also be detected. Repeated clogging of the HPLC column as a result of impurities usually present in considerable amounts in fermentation samples is another reason for using TLC for analysis of microbial media.

Absztrakt

A növényi alapú étrend egy régi-új táplálkozási irányzat. Jelen összefoglaló tanulmányban történeti alapokból kiindulva kívánjuk ismertetni ezt a napjainkban egyre népszerűbb táplálkozási törekvést. Célunk bemutatni a növényi alapú táplálkozást mint primer prevenciós eszközt. Irodalmi adatok alapján kritikusan elemezzük a növényi alapú étrendhez kapcsolódó korábbi téves vélekedéseket (fehérje-, B12-vitamin-, folsav-, vasbevitel), és megfelelő fogalmi alapot teremtünk ezen étrend értelmezéséhez, amelynek gerincét a gyümölcsök, zöldségek, teljes kiőrlésű gabonák, hüvelyesek és az olajos magvak alkotják. Ismertetjük a növényi alapú étrend pozitív élettani és betegségkockázat-csökkentő hatását. Az elhízás talaján kialakuló metabolikus és szív-ér rendszert érintő betegségek nagyrészt megelőzhetőek lennének egy helyesen összeállított növényi alapú étrenddel. A daganatos megbetegedésben szenvedő betegek esetén az állati eredetű élelmiszerek háttérbe szorítása a növényi alapúakkal szemben is igazoltan pozitív hatású. Áttekintésünk bemutatja, hogy ez a fajta táplálkozás számos egészségügyi probléma esetén alkalmazható, és hosszú távon is fenntartható megoldást biztosít a legújabb kor egészségügyi kihívásaira. Orv.
