3-nitropropionic acid (3-NP), a substance used for modelling Huntington’s disease, was given to male Wistar rats in a single 20 mg/kg b.w. dose, and the resulting behavioral alterations in spontaneous locomotor activity were measured after 30 minutes. To detect the involvement of neurotransmitter systems in this immediate effect, the NMDA antagonist MK-801 (0.8 mg/kg); as well as an agonist, quinpirole (QP, 5 mg/kg) and an antagonist, sulpiride (SP, 80 mg/kg) of the dopamine D2 receptors, were given before 3-NP to separate groups of rats. Controls were given saline. All substances were injected ip. 3-NP decreased the rats’ locomotor, especially vertical, activity, whereas local activity was increased. Based on the further changes of 3-NP effects in the combination groups it could be concluded that dopaminergic rather than glutamatergic mechanisms were possibly involved in the acute behavioral effect of 3-NP.

Concentrations of potentially toxic elements were determined in the soil solution of two soils (acidic sandy and slightly acidic clay loam) treated with phosphate rocks having high Cd content in a pot experiment. Relative concentrations characterizing the mobility of metals (expressed as soil solution concentrations in percentage of their “total” amounts in the phosphate rock-treated soil) decreased with increasing phosphate rock rates in the sandy soil. Mn@Sr>Cd@Co were the most, while Pb and Cr the least mobile elements. The relative concentrations in the clay loam soil were much lower than in the sandy soil and they practically remained constant with increasing phosphate rock rates. It was concluded that in the experimental time frame the environmental risk did not increase with the increase of phosphate rock rate.

Potassium uptake is the result of numerous simultaneous processes influencing the potassium dynamics in the rhizosphere.The presented research has focused on plant-soil interactions in the potassium supply of soil in the root environment of maize. It was assumed that: 1. roots promote the mobilization of K by the acidification of the rhizosphere soil, 2. roots increase wetting-drying cycles in their environment, and 3. soil total K content affects K release and fixation in the bulk of soil and the root environment.The promoting effect of root activity was detected on K release from soil when feldspar was added as K source to the root environment. A 2-unit reduction of soil pH multiplied K concentration in the soil solution, depending on the feldspar rate. Feldspar application significantly increased the solubility and release of potassium into the soil solution.The effect of pH reduction on the K concentration of soil solution was several magnitudes higher than that of the wetting-drying cycles both in the untreated and feldspar treated soils.Potassium uptake by maize over two generations greatly exceeded the exchangeable pool in the growing media. As a consequence of the exhaustive K uptake K release slowed down to the soil solution, as reflected in the H2O extractable K and ExK contents.Significant K fixation was detected after the K removal of maize in feldspar treated soils. On the contrary, in the treatments without plants increasing feldspar rates increased both H2O extractable K and ExK contents.One-term Langmuir equation, corrected with the originally sorbed amount of K, was fitted to measured data. The maximum amount of potassium adsorption (Kmax, mg∙kg−1) and the equilibrium constant (k) were calculated. The potassium buffering capacity was estimated at zero equilibrium concentration. Both K buffering capacity and the energy of K fixation were high for the rhizosphere soil. In rhizosphere soil samples the energy of K fixation was one magnitude higher as compared to the bulk soil and decreased substantially with feldspar addition. In soils without plants the k equilibrium constant did not change as the result of drying-wetting process only in the case of the 50% soil/feldspar mixture.In the liquid phase of the soil without feldspar application potassium concentration decreased in the one-year drying-wetting cycle, presumably it got into more strongly bounded forms in the low K status soil. In 50% feldspar enriched soil samples potassium concentration in the soil solution increased, likely as a consequence of a slow dissolution of the K content of feldspar.

Abstract

For the final quality of the part, metal forming trends are depending on improvements of friction and tribology. As a consequence, there is a trend in which tribology and friction are becoming increasingly important for correctly replicating the forming simulation of those parts. The objective of this work was to improve the forming simulation of an automotive shell part on AutoForm. The part was provided by a vehicle industry supplier. Enhanced Coulomb models consider a change on friction coefficient due to different factors, better approximating the description of friction to reality. For the current study, pressure and velocity dependent friction models were chosen as long as the combination of both. The virtual tribology was simulated using the software TriboForm. Velocity dependent friction model and the virtual tribology have shown similar results with expected lower coefficients of friction.

Abstract

Purpose

Proper handwashing helps prevent the spread of communicable diseases. The aim of our study was to analyse and compare children's knowledge and skills in hand hygiene before and after school interventions in order to evaluate the effectiveness of our peer education programme.

Materials/methods

In our longitudinal study, short- and long-term changes in the knowledge, hand-washing skills and health behaviour of 224 lower, upper and secondary school students were assessed. Our measurements were performed with a self-administered, anonymous questionnaire and the Semmelweis Scanner.

Results

As a result of the intervention, the proportion of correct answers increased significantly both in the short term and in the long run compared to the input measurements, but age differences did not disappear for most variables. There is a difference in the process of learning theoretical knowledge and practical skills. Areas not used for handwashing in the paediatric population are different from those described for adults in the literature. There was no significant difference between the mean scores of the right and left hands.

Conclusions

There was a significant positive change in both theory and practice of handwashing. In education, emphasis should also be put on long-term retention of theoretical knowledge in age-specific health promotion programmes within the paediatric population.

Mycoviruses are known to infect fungi of different habitats and life style. Some of them, like the Mushroom Virus X (MVX) complex, cause abnormal development of fruiting bodies and severe yield losses in mushroom cultivation. Most mycoviruses have a double-stranded RNA (dsRNA) genome, therefore dsRNA-detection is frequently used as a first step to identify virus infection. In relation with MVX 23 dsRNAs species have been described, occurring in variable number and combination in diseased mushrooms. The aim of our experiments was to find out whether dsRNA-immunoblotting can be used to detect dsRNA in small samples of cultivated A. bisporus varieties and of wild growing Agaricus species. We found that by immunoblotting, the same dsRNA species were detected in apparently healthy cultivated champignon fruiting bodies and in MVX-infected reference samples, respectively, as by conventional CF11 chromatography, but for immunoblotting a much smaller sample size was needed. In two out of three deformed fruit bodies of cultivated A. bisporus from Hungary we detected a 4.1 kbp dsRNA species which was also present in the MVX infected reference samples. Diverse and variable dsRNA patterns were observed in apparently healthy samples of 12 wild growing Agaricus species, indicating that extreme care should be taken when non-cultivated Agaricus is used for breeding new varieties. Non-sterile cultures and environmental mushroom specimens are fairly often mixed with parasitic and endofungal organisms, therefore, we also tested fungi isolated from mushroom cultures. Here again, 1–7 dsRNA species were found in extracts of Trichoderma and Dactylium isolates and of Mycogone-infected sporophores. Our results demonstrate clearly that dsRNAs from very different origins can be present in cultivated champignon and support the view that the MVX symptom-associated dsRNAs are probably of polyphyletic origin and do not represent one defined virus.

Expression of antibodies or antibody fragments in plants is a useful tool for producing active antibody derivatives for diagnostic or pharmaceutical purposes as well as for immunomodulation. We investigated the effect of cellular expression site on the stability and yield of double-stranded RNA (dsRNA)-specific single-chain Fv-fragments (scFv) in transgenic tobacco. Two antibodies (J2 and P6) belonging to the V23(J558) heavy chain variable gene family but differing in the light chain variable domain were used. scFvs were targeted to the cytoplasm - with or without anchoring them in the plasma membrane -, into the endoplasmic reticulum (ER) and to the apoplast. Although high mRNA concentrations were detected in all cases, scFv proteins accumulated only when scFvs were made ER-resident by appropriate signal sequences. When the ER retention signal was removed to allow scFv-secretion to the apoplast, no scFv-proteins were detected. Despite the strong homology of the VH-sequences of J2 and P6 antibodies, only P6 provided a stable scFv scaffold for intracytoplasmic expression. J2-scFv could not be stabilised either by adding a C-terminal stabilisation signal or by anchoring the protein on the cytoplasmic side of the plasma membrane (PM). It was found that dsRNA-specific J2-scFvs are active in vivo and enhance Potato Virus Y induced symptoms in infected tobacco. This is the first report describing the expression and biological effect of RNA-specific antibodies in plants.

There are several experimental models for the in vivo investigation of myocardial infarction (MI) in small (mouse, rat) and large animals (dog, pig, sheep and baboons). The application of large animal models raises ethical concerns, the design of experiments needs longer follow-up times, requiring proper breeding and housing conditions, therefore resulting in higher cost, than in vitro or small animal studies. On the other hand, the relevance of large animal models is very important, since they mostly resemble to human physiological and pathophysiological processes. The first main difference among MI models is the method of induction (open or closed chest, e.g. surgical or catheter based); the second main difference is the presence or absence of reperfusion. The former (i.e. reperfused MI) allows the investigation of reperfusion injury and new catheter based techniques during percutaneous coronary interventions, while the latter (i.e. nonreperfused MI) serves as a traditional coronary occlusion model, to test the effects of new pharmacological agents and biological therapies, as cell therapy. The reperfused and nonreperfused myocardial infarction has different outcomes, regarding left ventricular function, remodelling, subsequent heart failure, aneurysm formation and mortality. Our aim was to review the literature and report our findings regarding experimental MI models, regarding the differences among species, methods, reproducibility and interpretation.
