Several grapevine viruses were reported in Algeria and especially in grapevine germplasm collection, therefore it is a great challenge to free these varieties from virus infection before any breeding programs. Our study focused on the development of chemotherapy on autochthonous varieties collected in the grapevine germplasm collection of ITAFV. All these varieties were tested by DAS-ELISA and the presence of GLRaV-3 and GFLV was confirmed in all used samples for the sanitation. After 8 weeks of shoot tips in vitro culture in a modified M S medium containing ribavirin, DAS-ELISA test revealed that GLRaV-3 was completely eliminated and GFLV to a significant rate.

Abstract

A large amount of waste, especially the outer part of citrus fruits (peel), is generated after consuming the pulp and it remains unused. The valorisation of this waste by recovering its bioactive compounds seems interesting. The aim of this study was to find the optimal conditions using ultrasound-assisted extraction (UAE) that yield the highest carotenoid content and better antioxidant activity from Citrus reticulata Blanco peels.

Response surface methodology (RSM) through Box–Behnken experimental design was used to optimise the conditions for carotenoid extraction using UAE. Hexane concentration, temperature, and sonication time were selected as the main factors.

The results revealed that all independent variables affected the responses. The optimal UAE conditions for hexane concentration, temperature, and sonication time were 60.76%, 36.45 °C, and 37.32 min, respectively. The values of total carotenoid content (TCC) and total antioxidant activity (TAA) obtained by UAE were higher than those obtained by the maceration extraction method.

It can be concluded that the medium and extraction parameters, including hexane concentration, temperature, and sonication time, significantly influenced the recovery of carotenoids and antioxidant activity. The optimisation study allowed determining the appropriate conditions to maximise both responses. Compared to conventional maceration, the UAE method was superior and more efficient for extracting carotenoids from C. reticulata Blanco peels.

Pyrenophora teres f. teres (Ptt), causing net blotch in barley, is an important and frequently isolated leaf pathogen across the globe. The virulence spectrum of Ptt from North Africa including Morocco is poorly understood. Sixteen barley genotypes were challenged, at seedling stage, with 15 Ptt isolates that were collected from different agroecological zones of Morocco. The experiment was conducted in a factorial arrangement of treatments in a randomized complete block design with three replicates. The ANOVA revealed highly significant (P < 0.001) effects of genotype (G), isolate (I) and G×I interaction explaining 23.2, 62.5, and 13.9% of the variation, respectively. Therefore, the current study revealed highly diverse virulence pattern of Moroccan isolates. Furthermore, the results indicated that minor virulence of Ptt isolates dominated over virulence interaction. In addition, Taffa (6-rowed) and Aglou (2 rowed), had the highest level of resistance to Ptt, while Coast and Rabat071 were the most susceptible genotypes. Pt2, Pt7, Pt8 and Pt4 were being the most virulent isolates, while Pt10 and Pt11 were the least virulent isolates. The emergence of the new Ptt pathotypes, which were highly virulent to durable resistance in Rabat071 posed a risk of breaking down the currently deployed resistance to net blotch in Morocco. A careful evaluation and selection of Ptt isolates based on minor virulence pattern to barley genotypes is essential for successful barley breeding program for resistance to net blotch in Morocco.
