Summary

A simple and rapid capillary electrophoretic procedure for analysis of matrine and oxymatrine in Kushen medicinal preparations has been developed and optimized. Orthogonal design was used to optimize the separation and detection conditions for the two active components. Phosphate concentration, applied potential, organic modifier content, and buffer pH were selected as variable conditions. The optimized background electrolyte contained 70 mM sodium dihydrogen phosphate and 30% acetonitrile at pH 5.5; the separation potential was 20 kV. Each analysis was complete within 5 min. Regression equations revealed linear relationships (r > 0.999) between peak area and amount for each component. The detection limits were 1.29 μg mL−1 for matrine and 1.48 μg mL−1 for oxymatrine. The levels of the two active compounds in two kinds of traditional Chinese medicinal preparation were easily determined with recoveries of 96.57–106.26%. In addition, multiple linear regression and a non-linear model using a radial basis function neural network approach were constructed for prediction of the migration time of oxymatrine. The predicted results were in good agreement with the experimental values, indicating that a radial basis function neural network is a potential means of prediction of separation time in capillary electrophoresis.

Abstract  

Various techniques and methodologies of thermal conductivity measurement have been based on the determination of the rate of directional heat flow through a material having a unit temperature differential between its opposing faces. The constancy of the rate depends on the material density, its thermal resistance and the heat flow path itself. The last of these variables contributes most significantly to the true value of steady-state axial and radial heat dissipation depending on the magnitude of transient thermal diffusivity along these directions. The transient hot-wire technique is broadly used for absolute measurements of the thermal conductivity of fluids. Refinement of this method has resulted in a capability for accurate and simultaneous measurement of both thermal conductivity and thermal diffusivity together with the determination of the specific heat. However, these measurements, especially those for the thermal diffusivity, may be significantly influenced by fluid radiation. Recently developed corrections have been used to examine this assumption and rectify the influence of even weak fluid radiation. A thermal conductivity cell for measurement of the thermal properties of electrically conducting fluids has been developed and discussed.

Abstract  

The paper describes a new transient hot wire instrument which employs 25.4 μm diameter tantalum wire with an insulating tantalum pentoxide coating. This hot-wire cell with a thin insulating layer is suitable for measurement of the thermal conductivity and the thermal diffusivity of electrically conducting and polar liquids. This instrument has been used for experimental measurement of the thermal conductivity and the thermal diffusivity of poly(acrylic acid) solution (50 mass%) in the temperature range of 299 to 368 K at atmospheric pressure. The thermal conductivity data is estimated to be accurate within ±4%. Thermal diffusivity measurements have a much higher uncertainty (±30%) and need further refinement.

Abstract  

The transient hot-wire technique is widely used for absolute measurements of the thermal conductivity of fluids. Refinement of this method has resulted in a capability for accurate and simultaneous measurement of both thermal conductivity and thermal diffusivity together with a determination of the specific heat. However, these measurements, especially those for the thermal diffusivity, may be significantly influenced by fluid radiation. The present work investigates the effect of fluid radiation on the measurements of the thermal conductivity of propane. Recently developed corrections have been used to examine this assumption and rectify the influence of even weak fluid radiation. Measurements at 372 K with a hot-wire instrument demonstrate the presence of radiation effects in both the liquid and vapor phase. The influence is much more pronounced in liquid propane at 15.5 MPa than in the vapor phase at 881.5 kPa. The technique employed to obtain radiation-free thermal conductivity measurements is described.

Abstract

The transient hot-wire method is considered the most accurate technique to measure the thermal conductivity of fluids. In this study, a transient hot wire instrument which employs 25.4-μm-diameter tantalum wire with an insulating tantalum pentoxide coating has been used. This hot-wire cell with a thin insulating layer is suitable for measurement of the thermal conductivity of electrically conducting and polar liquids. Measurements of the thermal conductivity of 50 wt% solution of PAA [poly (acrylic acid)] in water and PAA–Na in 50 wt% water are reported here. These measurements were obtained in the temperature range of 299–368 K at 1 atmospheric pressure. The measurement of thermal conductivity is estimated to be accurate within ±4%.

In this paper, the wind energy resource in China’s oilfields is analyzed, the difference between China’s energy consumption structure and that of the world in average is analyzed as well, and the application prospect of wind heating technology in China’s oilfields is discussed as an example to reform China’s energy consumption structure. It shows that it is possible to use wind energy as an appropriate heat resource or supplementary heat source in some oilfields to supply heat energy for oil heating and living, more room to improve the energy resource utilization in various fields in China remain. It is a benefit to both oilfield and society to improve energy saving and environments.

Abstract  

Molar heat capacities (C p,m) of aspirin were precisely measured with a small sample precision automated adiabatic calorimeter over the temperature range from 78 to 383 K. No phase transition was observed in this temperature region. The polynomial function of C p,m vs. T was established in the light of the low-temperature heat capacity measurements and least square fitting method. The corresponding function is as follows: for 78 K≤T≤383 K, C p,m/J mol-1 K-1=19.086X 4+15.951X 3-5.2548X 2+90.192X+176.65, [X=(T-230.50/152.5)]. The thermodynamic functions on the base of the reference temperature of 298.15 K, {ΔH TH 298.15} and {S T-S 298.15}, were derived. Combustion energy of aspirin (Δc U m) was determined by static bomb combustion calorimeter. Enthalpy of combustion (Δc H o m) and enthalpy of formation (Δf H o m) were derived through Δc U m as - (3945.262.63) kJ mol-1 and - (736.411.30) kJ mol-1, respectively.

This study aimed to clarify the genetic mechanisms behind wheat flour color. Flour colorrelated traits (L*, a*, and b*) and polyphenol oxidase (PPO) activity are important parameters that influence the end-use quality of wheat. Dissecting the genetic bases and exploring important chromosomal loci of these traits are extremely important for improving wheat quality. The diverse panel of 205 elite wheat varieties (lines) was genotyped using a highdensity Illumina iSelect 90K single-nucleotide polymorphisms (SNPs) assay to disclose the genetic mechanism of flour color-related traits and PPO activity. In 2 different environments and their mean values (MV), 28, 30, 24, and 12 marker-trait associations (MTAs) were identified for L*, a*, b* traits, and PPO activity, respectively. A single locus could explain from 5.52% to 20.01% of the phenotypic variation for all analyzed traits. Among them, 5 highly significant SNPs (P ≤ 0.0001), 11 stable SNPs (detected in all environments) and 25 multitrait MTAs were identified. Especially, BS00000020_51 showed pleiotropic effects on L*, a*, and b*, and was detected in all environments with the highest phenotypic contribution rates. Furthermore, this SNP was also found to be co-associated with wheat grain hardness, ash content, and pasting temperature of starch in previous studies. The identification of these significantly associated SNPs is helpful in revealing the genetic mechanisms of wheat colorrelated traits, and also provides a reference for follow-up molecular marker-assisted selection in wheat breeding.

Two hundred and ninety F9 recombinant inbred lines (RILs) derived from the bread wheat cultivar Gaocheng 8901 and the waxy wheat cultivar Nuomai 1 were used in determining the high-molecular-weight glutenin subunit (HMW-GS) and waxy protein subunit combinations and their effects on the dough quality and texture profile analysis (TPA) of cooked Chinese noodles. Seven alleles were detected at Glu-1 loci. There were two alleles found at each of the Wx-A1, Wx-B1 and Wx-D1 loci. Eight allelic combinations were observed for HMW-GS, LMW-GS and waxy proteins, respectively. Both the 1/7+8/5+10 and 1/7+8/5+12 combinations contributed to dough elasticity, and the 1/7+8/5+10 combination also provided better TPA characteristics. Compared to Wx protein, HMW-GS was more important on dough alveogram properties. LMW-GS significantly affected springiness and cohesiveness; HMW-GS mainly affected the hardness; Wx×LMW-GS significantly affected the springiness, cohesiveness and chewiness; HMW-GS×Wx×LMW-GS mainly influenced the springiness and chewiness. But HMW-GS×LMW-GS only affected the spinginess. These indicated the TPA of noodles was significantly affected by the interactions between glutenin and Wx proteins.

Abstract  

Using TAM III multi-channel calorimetry combined with direct microorganism counting (bacteria, actinomycetes and fungi) under laboratory conditions, we determined the microbial population count, resistance and activity toward cadmium (Cd(II)) and hexavalent chromium (Cr(VI)) toxicity in soil. The thermokinetic parameters, which can represent soil microbial activity, were calculated from power-time curves of soil microbial activity obtained by microcalorimetric measurement. Simultaneous application of the two methods showed that growth rate constant (k), peak-heat output power (P max) and the number of living microorganisms decreased with increasing concentration of Cd and Cr. The accumulation of Cr on E. coli was conducted by HPLC-ICP-MS. Cr6+ accumulation by Escherichia coli was increased steadily with increasing Cr6+ concentration. The results revealed that the change in some thermo-kinetic parameters could have good corresponding relationship with metal accumulation. Our work also suggests that microcalorimetry is a fast, simple, more sensitive, on-line and in vitro method that can be easily performed to study the toxicity of different species of heavy metals on microorganism compared to other biological methods, and can combine with other analytic methods to study the interaction mechanism between environmental toxicants and microbes.
