Summary  

We discuss the relationship between two different sequential connectedness, and prove that sequential connectedness is countably multiplicative.

Spermatogonial stem cells (SSCs) are defined by unique properties like other stem cells. However, there are two major challenges: long-term cultivation of normal SSCs into stable cell lines and maintaining the SSCs as undifferentiated and capable of self-renewal. Here, we compared different culture methods for mouse SSCs isolated and cultured from testicular tissue. We found that human amniotic epithelial cells (hAECs) can behave as feeder cells, allowing mouse SSCs to maintain a high level of alkaline phosphatase (AP) activity when cultured long-term. Also, we observed that expression of Nanog, Oct-4 and other important stem cells markers were higher in mouse SSCs cultured on hAECs compared to those cultured on MEF or without any feeder cells. Furthermore, we demonstrated that the CpG islands of the Nanog and Oct-4 promoters were hypomethylated in cells cultured on hAECs. In addition, mouse SSCs cultured on hAECs exhibited higher levels of H3AC and H3K4Me3 in the Nanog and Oct-4 promoters than those cultured on MEF or without feeder cells. Taken together, these results suggest that the hAECinduced epigenetic modifications at the Nanog and Oct-4 locus could be a key mechanism for maintaining mouse SSCs in an undifferentiated state capable of self-renewal.

Abstract  

Multitracer technique was used to study the uptake and distribution of some relatively long half-life radionuclides Be, Na, Mn, Co, Sc to growing cucumber (Cucumis Sativus L.) with two different treatments. In Hoagland solution, only 54Mn and 60Co accumulated in the every part of plants. 54Mn, 60Co and other radionuclides were absorbed in distilled water. The results indicate that there were major differences in the accumulation of trace elements between the two different treatments.

To understand the genetic diversity of porcine reproductive and respiratory syndrome virus (PRRSV) in South China, we collected 231 clinical samples from pigs with suspected PRRSV infection in Guangdong between 2007 and 2009. We found that 74 of 231 samples were positive by RT-PCR. The PCR products of the ORF5 gene of 35 isolates from different farms were sequenced and their DNA sequences were compared to 23 other PRRSV isolates in the GenBank. We found that the nucleotide similarity among all South China isolates ranged from 87.6% to 100%, and all belonged to the North American genotype. Most of them were classified into subgenotype I, but the rest mapped to subgenotypes III, V or VI. Those in subgenotypes I and III were found to be highly variable in the primary neutralising epitope (PNE) with a specific amino acid mutation (F39/L39→I39), and a few isolates in subgenotypes I and III isolates also had a mutation at L41 (L41→S41). PRRSV isolates in subgenotypes III, V and VI had less potential glycosylation sites than those in subgenotype I. Our data contribute to the understanding of molecular variation of PRRSV in South China.

Abstract  

A total of 178 aerosol samples in two size fractions, PM10-2.5 (coarse) and PM2.5 (fine), were collected on nucleopore films using a Gent stacked filter unit sampler at the Graduate School of China Nuclear Industrial Group during May 2007 to November of 2009. Black carbon was determined by a reflectometer. A total of 16 elements, Mg, Al, S, Si, P, Cl, Ca, K, Ti, Mn, Fe, Ni, Cu, Zn, As and Pb, were determined by Particle Induced X-ray Emission. Mg, Al, Ca, K and Mn were also determined by Neutron Activation Analysis. Concentrations of all these elements were used to identify possible pollution sources and directions of the airborne particulate matter by means of softwares PMF and CPF. Some extraordinary events, such as sandstorms, firework and transboundary fire smoke were pinpointed by a combination of time series of multielement, relevant meteorological data and softwares Wind rose, Hysplit, and Google earth.
