Abstract
A thermodynamic study on the interaction of bovine carbonic anhydrase II (CAII) with nickel ions was performed by using isothermal titration calorimetry (ITC) at 27 °C in Tris buffer solution at pH = 7.5. The enthalpies of Ni2+ + CAII interaction are reported and analysed in terms of the new solvation theory. It was indicated that there are three identical and non-cooperative sites for Ni2+. The binding of a nickle ion is exothermic with dissociation equilibrium constants of 81.306 and 99.126 μM at 27°C and 37°C, respectively. The binding of nickel ions can cause some changes in the stability of the enzyme at low and high Ni2+ concentrations.
Considering that the excessive usage of vitamin E causes hypervitaminosis and thus reduces blood erythrocyte concentrations, therefore it is worth studying how its pharmacological dosage affects the activity of carbonic anhydrase (CA) enzyme found in erythrocytes of rainbow trout (Oncorhynchus mykiss) in vitro and in vivo. Vitamin E inhibited CA enzyme and the IC50 value of the vitamin was 0.039 mM in vitro. Similarly, it was seen that vitamin E inhibited CA enzyme activity after the first hour following vitamin E injections in vivo. The activities of CA in groups of trout given vitamin E injection were measured at 1, 3 and 5 h and the corresponding activities were found to be 772.7 ± 290.5 (P < 0.05), 1286.4 ± 378.2 and 1005.7 ± 436.1 enzyme units (EU) g Hb-1. The difference over the control was significant (P < 0.05) in the first hour and insignificant at 3 and 5 h (P ? 0.05). The activity of CA in the control, which did not contain vitamin E, was determined as 1597.7 ± 429.0 EU g Hb-1.

Abstract
A novel method for the characterization of metal containing biological compounds has been developed which utilizes both nuclear activation [e.g.,12C(3He, α)11C] and atomic activations (X-ray emission) induced by 8 MeV3He bombardment. Detection limits for carbon (≈1 μg), oxygen (≈1μg), and metals (1 to 10 ng for elements between P and Zn) were obtained under routine experimental conditions. The metal stoichiometry of six different compounds (alkaline phosphatase, amylase, carbonic anhydrase, diamine oxidase, my oglobin, vitamin B12) were determined with typical relative precisions of ±25% for a 40 μg sample. A comparison of these ratios with carbon and metal measurements obtained by other methods showed a relative accuracy of 1 to 20%.
